A novel protein having inhibitory activity on the protease activity of hepatocyte growth factor (HGF) activator was purified and isolated, and its molecular weight (ca. 40,000 daltons) and its N-terminal and partial amino acid sequences were determined. A gene coding for the protein was cloned, and the gene DNA was incorporated into a vector, for transforming host cells. Cultivation of the transformant gave the desired protein. The protein can be used as an in vivo or in vitro control factor for HGF or HGF activator. It is also useful as an antigen to be used in producing an antibody to be used as means for kinetic studies of the protein, or as a standard in assay systems therefor.

In the manufacture of a substrate with a hydrophobic film used for a reference electrode of an ion sensor or the like, a hydrophobic film is formed on a substrate by irradiating a target consisting of a hydrophobic compound with a neutral atom beam and thereby effecting sputtering. The apparatus for effecting the sputtering comprises a target base disposed in a vacuum chamber, an atom beam gun for irradiating a target on the target base with a neutral beam, a substrate base and a shutter for controlling the passage of sputtered particles. A thin film that is manufactured is suitable for an ion sensor, such as an ISFET or the like or an enzyme sensor.

A reference electrode for generating a reference potential on an ion sensor is disclosed. The reference electrode has a lamination film covering a surface of an electrically conductive substrate and formed by alternately laminating silver halide thin films and hydrophobic resin thin films.

An enzyme sensor is disclosed, which has a two-layer fixed film structure consisting of a reduction function layer including an electron movement medium and di-valent iron ions and an enzyme-fixed layer covering said reduction function layer, and in which hydrogen peroxide generated through decomposition of glucose or the like in the enzyme-fixed layer is reduced by electron movement medium and di-valent iron ions in the reduction function layer. Thus, the construction of enzyme sensor is simplified, measurement is extremely facilitated, and the measurement time is reduced. As a different principle of enzyme sensor, an electrically conductive substrate of conductive carbon or the like is covered with an oxygen-reducing catalytic function layer and enzyme layer. Thus, there is no problem of contamination of enzyme sensor, and it is possible to readily manufacture and miniaturize enzyme sensors.

An electrochemical cell is formed by immersing an electrochemical ion sensor and a reference electrode into a solution of interest, the electrochemical ion sensor exhibiting ion selectivity with respect to an ion of interest and having a high electrode resistance. A current which flows through the electrochemical cell is measured while the potential of the electrochemical ion sensor is held to a constant potential with respect to the reference electrode. The current so obtained are proportional to the logarithm value of the concentration of the ion.
