A nucleic acid sequence measuring method includes measuring fluorescence from the nucleic acid sequence measuring device supplied with a sample solution. The device includes a fluorescent probe added with a fluorescent molecule, and a quenching probe added with a quenching substance. The fluorescent probe and/or the quenching probe has a detection part detecting a predetermined nucleic acid sequence. Fluorescence from the fluorescent molecule is quenched by the quenching substance coupled with the fluorescent molecule when the hybridization between the detection target nucleic acid and the detection part has not occurred, and fluorescence is emitted from the fluorescent molecule separated from the quenching substance when the hybridization has occurred.

There are provided a capsule and a chemical reaction cartridge capable of properly handling reagents and so forth. The capsule is formed by laminating two dome-shaped films together at the peripheries thereof. The films are formed by subjecting heat sealable films, to which aluminum vapor deposition is applied, to a drawing process. The films are made of a material (easy-peel material), which can be varied in seal strength by a heating temperature and are thermally welded together at an adhesion area at the peripheries thereof. Thereafter, contents such as reagents and so forth are filled up inside the capsule via a filling section.

The present invention makes it possible to realize a chemical reaction cartridge, which enables prescribed protocols to be achieved easily without differences among operators. The cartridge is sealed and disposable, and has a safe structure against viruses or dangerous drugs. This cartridge is intended to be used for performing chemical reaction of samples. The cartridge comprises a container formed of a rigid substrate and an elastic body, wherein two or more chambers linked or arranged to be able to be linked through flow paths are formed in the container. The flow paths, the chambers or both are partially closed by applying an external force to the elastic body from outside the container, thereby being able to move or block fluid substance in said flow paths or chambers.

A chemical reaction cartridge includes a vessel having at least one part made of an elastic body, in which a plurality of chambers formed in the vessel so as to be connected or arranged connectably through a flow path, and an external force is applied to the elastic body from an outside of the vessel to move a fluid substance in the flow path or the chambers or in both the flow path and the chambers so as to perform a chemical reaction. At least a chamber into which the fluid substance is flowed has an air-release path which releases air from the chamber.

A method for measuring a fluorescent light amount, whereby a specimen is irradiated with excitation light, and fluorescent light generated from the specimen is measured by a light-receiving element via of a receiving optical system, the method includes the steps of irradiating the specimen with excitation light having a light-amount value per specified unit area traceable to the national standard, and designated on the surface of the specimen beforehand, causing the light-receiving element to measure the fluorescent light generated from the specimen, and computing a fluorescent light-amount as measured by the light-receiving element of the receiving optical system on the basis of an excitation light-amount per the specified unit area, optical coefficients of the receiving optical system, and the acceptance coefficient of the light-receiving element, thereby working out a light amount value per the unit area traceable to the national standard.

Method for measuring fluorescent light amount or light absorption amount and device for measuring the same

A biochip reader wherein spectroscopic information of a sample under analysis is arranged in spaces between images of the sample arranged on a biochip. The reader comprises a confocal microscope and the biochip comprises a transparent substrate to allow passage of the excitation light and fluorescent light from the sample with the excitation light being applied from the side opposite that on which the samples are arranged so that noise from dust and the like is avoided by the transmitted light avoiding contact with the dust. Another aspect is an electrophoresis system wherein different coloring material are used for each of a variety of target substances, so that the same lane and area are utilizable to concurrently detect a polychrome fluorescent pattern of the different targets. A confocal scanner or fluorescence imaging system is used with a plurality of filters to detect the multi-colored fluorescences of the target substance. Advantageously, in the biochip reader, a lower S/N ration is obtained together with lower cost; and in the electrophoresis system, concurrent detection of multiple polychromatic fluorescence patters is attained.

There is provided a molecularity measurement instrument capable of working out the number of molecules in a sample by comparing a measured value of a light quantity with a theoretical light quantity per a single molecule, and a molecularity measurement method using the same. The molecularity of the sample is quantitatively estimated on the basis of a light quantity having correlation with the molecularity. The molecularity measurement method comprises the step of working out a theoretical light quantity per a single molecule, the step of measuring a light quantity of the sample by use of an image detector, and the step of working out the molecularity of the sample on the basis of a ratio of the light quantity of the sample to the theoretical light quantity as worked out.

The present invention is characterized by the following points: In a biochip reader used for reading a measurement sample image by light beam irradiation, a correction method for the distribution of quantity of light which is devised to remove the influence of shading for the whole image and such a biochip reader can be realized by correcting non-uniformity in said quantity of light in light beam irradiation by dividing the quantities of light of pixels in a measured image obtained from the measurement of a measurement sample by a distribution of quantity of light in an image obtained from the measurement of a uniform fluorescent plate that presents a uniform fluorescent light distribution, the positions of pixels in the measured image being correspondent to those in the image obtained through the above uniform fluorescent plate measurement.

Correction method for the distribution of quantity of light and biochip-reader

A cartridge adaptable to various measurement forms is provided. A sample is injected into a well via an injection path. When a roller while kept pressed into contact with the cartridge is rotated rightward, the sample held in the well, and liquid solvent held in a well are caused to reach a well via flow paths, respectively, thereby mixing the sample with the liquid solvent. A mixed liquid is branched into flow paths, respectively, to thereby reach wells, respectively, and reagents pre-held in wells, respectively, are caused to flow into the wells, respectively. Next, temperatures in the wells, respectively, are controlled, thereby executing DNA amplification. Then, PCR by-products as amplified in the wells, respectively, are transferred as far as a position in the vicinity of an electrode via flow paths. Subsequently, a voltage is applied across the electrode as a negative electrode, and an electrode as a positive electrode via a drawing electrode and a drawing electrode, to thereby cause the PCR by-products to undergo electrophoresis.

The present invention describes a disposable chemical reaction cartridge comprising a container formed of a rigid substrate and an elastic body, wherein two or more chambers are linked or configured to be linked through flow paths, and wherein the flow paths, the chambers or both, can be partially closed by applying an external force to the elastic body from outside of the container, to move or block fluids in the flow paths or chambers. The cartridge has a structure protected against viruses or dangerous drugs and facilitates foolproof implementation of prescribed protocols for the samples to be tested.

In a cartridge for chemical reaction, there are provided an inner part in which a chemical reaction is caused by feeding a liquid in accordance with a deformation when an external force is applied, and a plurality of passages that are connected with each other, and the passages can be switched between an opened state and a closed state by the external force. Since the plurality of passages are connected together in the cartridge, the cartridge can be applied to various uses. The plurality of passages may be connected together in the form of a network. Wells may be provided at the junctions of the passages. When the external force is applied on the well, the liquid may be pushed out of the well through the passages connected to the well. A passage may be provided for introducing a sample into the cartridge from an external part of the cartridge.
