The invention provides a method of producing a protein multimer-nucleic acid complex comprising a protein multimer and any one target component of the protein multimer. A nucleic acid encoding the target component is subjected to in vitro translation to provide a translation product containing a target component-nucleic acid complex of the target component and the nucleic acid encoding the target component. A nucleic acid encoding a non-target component constituting the protein multimer together with the target component is translated into the non-target component by adding the nucleic acid encoding the non-target component to the previously provided translation product to provide the non-target component. The non-target component then is associated with the target component contained in the target component-nucleic acid complex to form a protein multimer, thus affording the protein multimer-nucleic acid complex of the protein multimer and the nucleic acid encoding the target component.

The polynucleotide construct of (1) or (2) below is used to perform ribosome display, CIS display and/or mRNA display in order to screen a Fab against an antigen of interest: (1) a polynucleotide construct which monocistronically comprises a ribosome-binding sequence, Fab first chain-coding sequence, linker peptide-coding sequence, Fab second chain-coding sequence and scaffold-coding sequence in this order, and further comprises at its 3'-end a structure necessary for maintaining a complex with the Fab encoded by itself; and (2) a polynucleotide construct which comprises a Fab first chain-expressing cistron and a Fab second chain-expressing cistron each containing a ribosome-binding sequence, a Fab first chain-coding sequence or Fab second chain-coding sequence, and a scaffold-coding sequence in this order, the first Fab-expressing cistron further comprising at its 3&#x2032;-end a ribosome stall sequence, said Fab second chain-expressing cistron further comprising at its 3&#x2032;-end a structure necessary for maintaining a complex with the Fab encoded by itself.

According to the present invention, a composition possessing cell-free protein synthesis activity with reduced contaminating lipopolysaccharide, and a method for producing a protein using the same are provided. When ribosome display is performed using the composition and method for protein production of the present invention, the background that is caused by non-specific binding is reduced, so that a nucleic acid that encodes the desired polypeptide can be selected with high accuracy and high efficiency.

The present invention provides a method of producing a maturated oligonucleotide library, including a step of obtaining a terminal-modified product of a maturation target oligonucleotide library, including adding a tag sequence to the 5' terminus of the maturation target oligonucleotide library and an arrest sequence, which stalls translation elongation on a ribosome, to the 3&#x2032; terminus of the maturation target oligonucleotide library, a step of transcribing the terminal-modified sequence product to give a transcript, and a step of in vitro translation for translating the transcript in vitro, wherein the maturation target oligonucleotide library is a random oligonucleotide library.

High-speed maturation method for an oligonucleotide library for the purpose of preparing a protein library

A heating resistor includes a conductive material, and an electrically insulating material made of a mixture of silicon carbide (SiC) and carbon (C) having a carbon content of not less than 3 wt % based on the mixture. The resistor is formed by sputtering of a target of the conductive material and the electrically insulating material having at least 5 wt % of carbon. The conductive material and the electrically insulating material may be respectively provided as separate targets. The conductive material is at least one metal selected from tantalum, niobium, titanium, tungsten, molybdenum, zirconium and chromium.

Heating resistor and method for making same

A heater element plated on a dielectric substrate through electroless plating process provides an excellent thermal head for thermally printing on a paper. Said heater element composes of nickel, one of phosphorus and boron, and some minor additive agent. That additive agent is one of tungsten and molybdenum by 2-10 weight %. Because of the addition of the additive agent, the life time and heat stress characteristics of a thermal head are considerably improved.

Thermal head

A thermal head for a thermal printer and/or a thermal ink transfer printer having a substrate, a heater layer and a conductive lead layer on the substrate in which a protection layer covering the heater layer has been improved. According to the present invention said protection layer is made of polyimide resin which includes some hard particle of filler of S.sub.i C with a weight ratio to the polyimide solid in the range between 1.1 and 3.2. Since the present protection layer is provided with low curing temperature, the substrate of the present thermal head may be polyimide resin which is not heat-proof. Because of filler in the protection layer, that protection layer is wear-proof, although polyimide layer itself is not wear-proof.
