An object of the present invention is to provide a biosensor comprising a hydrogel capable of immobilizing a physiologically active substance thereon, which can be produced conveniently by use of a safe material, and a method for producing the same. The present invention provides a method for producing a biosensor, which comprises bringing a polymer containing an activated carboxyl group into contact with a substrate surface coated with an organic layer having an amino group to thereby bind the polymer to the organic layer.

Method for producing a biosensor

One object of the present invention is to provide a biosensor and a production method therefor, by which hydrogel that enables immobilization of a physiologically active substance can be conveniently produced using safe raw materials. The present invention provides a biosensor which comprises a substrate having a metal layer on its surface, wherein a hydrophilic polymer having a reactive functional group capable of reacting with a hydroxyl group or an amino group of a physiologically active substance is bound to the metal layer directly or indirectly via an intermediate layer.

Thus, the present invention provides a method for producing a biosensor comprising a substrate coated with a hydrophilic polymer, which comprises allowing a compound generating a reactive group as a result of external stimulus to bind to the surface of a substrate, and then giving external stimulus in a state where a hydrophilic polymer capable of forming a covalent bond with said reactive group is allowed to come into contact with the substrate, so that said hydrophilic polymer is allowed to bind to the substrate surface via a covalent bond with said reactive group.

An object of the present invention is to provide a biochemical instrument having a surface that can simultaneously inhibit adsorption of a biopolymer and that of a hydrophobic low-molecular-weight compound. The present invention provides a biochemical instrument comprising a region that inhibits adsorption of a hydrophobic low-molecular-weight compound and a region that inhibits adsorption of a biopolymer on the surface of a water-insoluble material.

A method of manufacturing a polymer film having a microphase-separated morphology includes a film-forming step in which a solution containing a block copolymer having two or more types of polymer chains which are mutually incompatible and are bonded to each other, a first solvent and a second solvent having a higher boiling point by at least 15&#xb0; C. than the first solvent is applied onto a substrate and the first solvent is removed to form a film; a heating step in which the film is heated at a temperature which is higher than the boiling point of the first solvent but is lower than the boiling point of the second solvent to make the film formed have a microphase-separated morphology; and a solvent removal step in which the second solvent is removed from the film having the microphase-separated morphology to form the polymer film having the microphase-separated morphology.

An object of the present invention is to provide a biosensor and a method for immobilizing a physiologically active substance, by which preconcentration effects can be obtained at a pH that is equivalent to or higher than the isoelectric point of the physiologically active substance and the physiologically active substance can be covalently bound to the surface. A method for purifying bioactive substances includes the steps of: causing a bioactive substance having histidine units to contact media, each constituted by a substrate, ligands which are physically attached to the surface of the substrate, and Cu(II) or Fe(II) metal ions which are covalently bonded to the ligands; causing the bioactive substance to covalently bond with the metal ions via the histidine units; and washing the media with an amount of 1 nmol/L to 10 mmol/L imidazole derivative solution 60 times the volume of the media or greater.
