The present invention relates to immunogenic immune complexes, related compositions, and related methods.

Immune complex

The present disclosure provides methods related to determining the level of afucosylated Fc glycans in IgG antibodies in a biological sample from a subject. This level can be used in methods aimed at monitoring and/or treating subject suffering from an acute flavivirus infection and/or who are at risk for progression to clinically significant infection or disease. This level can also be used in a vaccination method to ensure that those who receive a flavivirus vaccine have a reduced risk of reacting to the vaccine be developing clinically significant infection or disease. The disclosure also provides treatment methods based on inhibiting Fc&#x3b3;RIIA or Fc&#x3b3;RIIIA receptor signaling. Also provided are novel cell lines that are useful in measuring afucosylated Fc glycans.

Measurement of afucosylated IgG Fc glycans and related vaccination methods

The present invention relates to immunogenic immune complexes, related compositions, and related methods. This invention addresses the above-described un-met need by providing an immune complex (IC) or immune complexes (ICs), which take advantages of Type II Fc receptor (FcR) interactions and signaling to elicit broader and more potent protective immune responses. In one aspect, the invention provides an immune complex comprising an antigenic agent and an isolated protein. The protein comprises an IgG Fc region and is capable of binding to a Type II Fc receptor. Preferably, the protein is an antibody or an Fc region-containing fragment thereof.

Provided herein are polypeptides comprising portions of the influenza virus hemagglutinin, compositions comprising such polypeptides that can be used as immunogens in vaccines and methods of their use to generate an immune response against multiple influenza subtypes in a subject.

Vaccines for use in the prophylaxis and treatment of influenza virus disease

Provided herein are methods of producing neutralizing monoclonal antibodies, by cyclical immunization, that cross-react with strains of Influenza virus of the same subtype or different subtypes. Also provided herein are compositions comprising such antibodies and methods of using such antibodies to diagnose, prevent or treat Influenza virus disease.
