The present description is related to the field of protein production. It introduces novel host cells with low protease activity, a novel protease regulator, its use in expression systems and protein production, and a method of producing host cells for protein production.

Methods for controlling protease production

The present invention relates to polypeptides having cellulase activity, in particular to variants derived from the 20K-cellulase enzyme. The invention discloses a number of amino acid residue positions important for the properties of the cellulase enzyme and thereby for the stability and/or performance thereof. The novel variants have improved stability compared to the parental cellulase. In particular, the novel variants have good performance in an antigreying application and excellent stability in the presence of a protease in several detergent compositions even in long-term experiments.

Variants of fungal cellulase

The present invention is related to a fungal serine protease enzyme useful in modification, degradation or removal of proteinaceous material, which enzyme comprises an amino acid sequence of the mature Tr Prb1 enzyme having an amino acid sequence of SEQ ID NO: 10 or a variant thereof having similar activity. The serine protease is obtainable from. Also disclosed are nucleic acid sequences encoding said protease, such as plasmid pALK2650 comprising the nucleotide sequence SEQ ID NO: 10 of the full length enzyme deposited inRF8052 under accession number DSM 22635. Said protease is useful as an enzyme preparation applicable in detergent compositions and for treating fibers, for treating wool, for treating hair, for treating leather, for treating food or feed, or for any applications involving modification, degradation or removal of proteinaceous material at low or moderate temperature ranges.
