Images of two-dimensional chromatograms or other sample arrays are formed on a scanning instrument that utilizes a line of illumination light that sweeps the length of the array either by moving across the array or by the array moving relative to the light, in either case scanning the entire two-dimensional array with a unidirectional pass of the moving component. The use of a CCD equipped with time delay integration allows the instrument to form an enhanced image.

Imaging of two-dimensional arrays

A method for the simultaneous and quantitative, flow cytometric analysis of nucleated red blood cells (NRBC), white blood cells (WBC), damaged white blood cells and a white blood cell subclass differential (WBC/Diff) is provided. The method includes mixing an aliquot of a whole blood sample with a reagent system comprising a red blood cell (RBC) lysing component which lyses RBCs and NRBCs while minimizing damage to WBC cellular membranes and a membrane-impermeant nucleic acid stain which stains exposed NRBC nuclei and damaged WBCs, subjecting the stained aliquot to flow cytometric light measurements, obtaining at least one signal for the parameters of fluorescence (FL) and scattered light at a first and a second range of scatter angles, qualifying the obtained signals using AND/OR logic wherein to be qualified, a signal must be greater than the second scatter signal threshold AND also greater than either the first scatter signal threshold OR the FL threshold, constructing a three-dimensional plot of qualified intensity signals of fluorescence and scattered light from the detected signals, and differentiating the NRBC, WBC, damaged WBC and WBC/Diff from the constructed three-dimensional plot and determining the number of cells of each.

Method for simultaneous analysis of cell viability, nucleated red blood

A method of performing a first analysis and a second analysis on a single blood sample obtained with a single blood draw from a patient with an automated analyzer includes the steps of supplying the single blood sample obtained with the single blood draw from the patient to the automated analyzer. A memory on the automated analyzer containing a software routine is automatically accessed. The software routine is useful to adapt an optical system on the automated analyzer to correspond to the first analysis and the second analysis. The optical system on the automated analyzer is automatically adapted with the software routine in real time to correspond to the first analysis. The first analysis is automatically performed with the automated analyzer. The optical system on the automated analyzer is automatically adapted with the software routine in real time to correspond to the second analysis. The second analysis is automatically performed with the automated analyzer.

Fully automated analysis method with optical system for blood cell

A pseudo telecentric flow cytometric optical system for the simultaneous detection of several characteristics of particles suspended in a flowing medium. The system is made up of a flowcell through which the particles pass substantially one particle at a time, an optical system for directing light from a light source onto the flowing particles in the flowcell, a side angle optical collection system for receiving light from the flowing particles and for directing the light to one or more of a first set of detectors, and a forward angle collection system for receiving light from the flowing particles and for directing the light to one or more of a second set of detectors. The side angle optical collection system can include a condenser lens for directing light toward the first set of detectors with an exit pupil of the condenser lens located at the back focal plane of the condenser lens; a photosensitive surface of one or more of the first set of detectors is located at conjugate points of the back focal plane of the condenser lens such that an image of the exit pupil is positioned at the photosensitive surface of one or more of the first set of detectors. The forward angle optical collection system can include a collecting lens for directing light toward the second set of detectors where the collecting lens exit pupil is located in the back focal plane of the collecting lens and a photosensitive surface of one or more of the second set of detectors is located at the back focal plane of the collecting lens.

Pseudo telecentric optical design for flow cytometric blood cell analyzer

A method and a device for the simultaneous and quantitative, flow cytometric analysis of nucleated red blood cells (NRBC) and white blood cells (WBC) in a whole blood sample. The method includes the lysis of red blood cells (RBC) and NRBC cytoplasm from an aliquot of a whole blood sample to eliminate RBC and to expose the NRBC to a vital nuclear stain and the inhibition of the permeation of the nuclear stain into the WBC, subjecting the stained aliquot to flow cytometric light measurements, obtaining at least one signal for parameters including scattered light at a first and a second range of scatter angles and fluorescence (FL), qualifying the obtained signals by using the combination logic wherein a qualified signal must be greater than the second scatter signal threshold, while at the same time it must be greater than either the first scatter signal threshold or the FL threshold {[(first scatter angle signal OR FL signals) AND second scatter angle signal]}, constructing a three-dimensional plot of qualified intensity signals of fluorescence and scattered light from the detected signals, and differentiating the NRBC and WBC from the constructed three-dimensional plot and determining the number of cells of each.

Method for rapid and simultaneous analysis of nucleated red blood cells

This invention encompasses a work station for conducting assays which comprises filtering assembly, a slide reading assembly, and a slide with a porous filter membrane wherein the slide has a means for aligning the slide within the filter assembly to filter reagents on predetermined portions of the porous filter membrane and also to position the slide in the slide reading assembly so that detection will occur in those same predetermined locations.
