Methods for construction of infectious PAV genomes by homologous recombination in procaryotic cells are provided. Recombinant PAV viruses are obtained by transfection of mammalian cells with recombinant PAV genomes. The PAV-3 genome can be used as a vector for the expression of heterologous nucleotide sequences, for example, for the preparation and administration of subunit vaccines to swine or other mammals. In addition, PAV-3 vectors can be used for gene therapy and expression of heterologous polypeptides. PAV-3 genome sequences can also be used for diagnostic purposes, to detect the presence of PAV-3 DNA in a subject or biological sample.

The present invention relates to the characterization of the porcine adenovirus E1 and E4 regions. The complete nucleotide sequence of the genome of porcine adenovirus type 3 (PAV-3), providing the characterization of the PAV3 E1 region, is described herein. The present invention also provides helper vectors comprising a BAV sequence essential for encapsidation which is used in a helper virus for propagating recombinant adenovirus. The present invention relates to methods for culturing circovirus and in particular, porcine circovirus. The present invention provides compositions and methods for culturing porcine circovirus in mammalian cells expressing mammalian adenovirus E1 functional protein.

The invention provides cell lines capable of supporting the replication of a defective recombinant virus vector. In one aspect, bovine cell lines expressing adenovirus E1 functions are provided. The cell lines are useful for the propagation of adenovirus vectors with mutations and/or deletions in E1 and other essential regions of the adenovirus genome.

Bovine cells expressing adenovirus essential functions for propagation of recombinant adenoviral vectors

The present invention provides novel recombinant mammalian cell lines expressing bovine adenovirus functions. In one embodiment, mammalian cell lines stably transformed with bovine adenovirus E1 sequences express bovine adenovirus E1 gene products and are useful for the replication of bovine adenovirus vectors having deletions and/or insertions in the E1 region. Such vector/cell line combinations are useful for synthesis of recombinant polypeptides, preparation of subunit vaccines, and in gene therapy.
