A method of detecting a ligase expressing micro-organism in a sample comprises steps of treating the sample under conditions that inhibit the activity of ATP-dependent ligase from mammalian cells but which do not inhibit the activity of the microbial ligases, contacting the sample or a portion of the sample with a nucleic acid molecule which acts as a substrate for ligase activity in the sample, incubating the thus contacted sample under conditions suitable for ligase activity; and specifically determining the presence and/or the amount of a ligated nucleic acid molecule resulting from the action of the ligase on the substrate nucleic acid molecule to indicate the presence of the ligase expressing micro-organism. The micro-organism may be a fungus or a bacterium or both. High pH conditions may be employed to inactivate mammalian ligases. Related kits are described.

Aggregating proteins such as IgG, are bound selectively in the presence of the non-aggregated protein using a polyionic binding agent such as dextran sulphate or pentosan (anionic), or polyamine compounds such as pDADMAC (cationic) under selective binding conditions including the use of n-lauroylsarcosine at mildly alkaline pH, and may then be assayed.

Micro-organisms, including fungi, viruses and bacteria such asand/or fragments of micro-organisms such as cell wall components present in an aqueous liquid are captured to a solid surface by adding to the liquid a sufficient quantity of a water soluble polymer in the presence of the solid surface to displace the micro-organisms and/or fragments from the liquid to the solid surface. The surface may be provided by a bead. The water soluble polymer may be polyethyleneglycol or polyvinylpyrrolidone.

An improved method of detecting an enzyme in a sample, which enzyme is capable of adding or removing a chemical moiety to or from a nucleic acid molecule, thereby conferring the nucleic acid molecule with the ability to be extended to generate a novel detectable nucleic acid molecule, comprises the steps of allowing the sample to be tested for the presence of the enzyme to interact with the nucleic acid molecule; and testing for interaction of the enzyme with the nucleic acid molecule by detecting the novel nucleic acid molecule generated only in the presence of the enzyme. The preferred enzyme is a phosphatase. The methods have a number of applications, for example in enhancing the sensitivity of immunoassays, for detecting pathogen associated phosphatase, for diagnosing certain conditions and for detecting specific contaminants in a sample.

Methods and kits for detecting an enzyme capable of modifying a nucleic acid

A method of detecting a molecule associated with viability of one or more cells or organisms in a sample comprises the initial step of contacting the sample with an enzyme, which enzyme is capable of adding or removing a chemical moiety to or from a nucleic acid molecule in the presence of the molecule associated with viability of the of the one or more cells or organisms. This thereby generates a novel detectable nucleic acid molecule. The next step involves detecting the presence of the molecule associated with viability of the one or more cells or organisms by detecting the novel nucleic acid molecule generated only in the presence of the molecule associated with viability of the one or more cells or organisms. A most preferred molecule associated with viability is ATP, although NAD may also be detected. A preferred enzyme for use in the methods is ligase. The method has numerous applications, in particular in monitoring viability of cells, toxicology testing and determining whether there is contamination in a sample or on a surface. Kits are also provided for carrying out the methods.

The present invention relates to a method for detecting a target material in a sample, which method comprises the steps of: a) exposing a sample, expected to contain that target material, to at least two viruses which are capable of binding directly or indirectly with that target material so as to form a virally bound target material and to endow the virally bound target material with a distinctive property; and b) cultivating the product from stage a in the presence of an indicator material to which the viruses carried by the virally bound target material attach so as to cause the indicator material to adopt the distinctive property of the virally bound target material; and c) monitoring the presence or otherwise of virally attached indicator material. Analytical method using multiple virus labelling

The present invention relates to a method for enhancing the time of response of an assay for a first bacterium, wherein: a) the first bacterium is exposed to infection by phage particles to which the first bacterium is permissive; b) the infected bacterium is treated to inactivate exogenous phage particles; c) the treated bacterium is cultivated in the presence of a second bacterium which is permissive to infection by the phage or its replicand and which has a doubling rate greater than the effective doubling rate of the first bacterium; and d) assessing the extent of plaque formation and/or of second bacterium growth in the cultivated second bacterium cells. The method can be used to assess the presence of first bacterium in a sample, notably where the first bacterium is a slow growing bacterium, such as , where the method enables an operator to detect the presence of low amounts of the bacterium in sample within days instead of weeks as required by conventional cultivation techniques. The invention can also be used to assess the effect of a drug or other treatment on a bacterium or on a virus. The invention also provides a diagnostic kit for use in the method of the invention.
