The present invention relates to the field of radiopharmaceuticals for in vivo imaging, in particular to automated methods for the preparation and purification ofF-labelled tau imaging radiotracers. Also provided are interchangeable cassettes useful in the methods, and the use of automated synthesizers and cassettes in the methods.

Radiolabelling method

The invention relates to a process for preparation of a compound of formula (I): Ris selected from Calkyl, Calkenyl, and Calkynyl;which comprises:(i) reaction with fluoride, suitably [F]fluoride, of a corresponding compound of formula (II): wherein Ris selected from hydrogen, Calkyl, Chaloalkyl, Caryl, Carylalkyl, &#x2014;(CHCHO)&#x2014;CHwherein q is an integer of from 1 to 10;Ris as defined for the compound of formula (I); andRis a leaving group. Certain novel precursors of formula (II) and radiopharmaceutical kits containing such precursors are also claimed.

Fluorination process

A diagnostic imaging agent useful for positron emission tomography (PET) imaging as well as to improved hardware for producing such imaging agents is described. Flutemetamol (F) Injection for the imaging of &#x3b2;-amyloid plaques in the brain and methods and devices for preparing same are described including the automated synthesis and purification of [F]flutemetamol by means of solid phase extraction (SPE).

Purification method

The invention relates to a process for preparation of radiopharmaceutical precursors, and in particular protected amino acid derivatives which are used as precursors for production of radiolabelled amino acids for use in in vivo imaging procedures such as positron emission tomography (PET). Particularly, the invention relates to a process for preparation of a precursor of the [F]-1-amino-3-fluorocyclobutanecarboxylic acid ([F] FACBC) PET agent, ensuring that the reaction efficiently goes to completion.

Precursor compounds and methods for making same

The present invention provides a PET tracer that has improved properties for imaging the peripheral benzodiazepine receptor (PBR) as compared with known such PET tracers. The present invention also provides a precursor compound useful in the preparation of the PET tracer of the invention and methods for the preparation of said precursor compound and said PET tracer. Also provided by the present invention is a radiopharmaceutical composition comprising the PET tracer of the invention. Methods for using the PET tracer and the radiopharmaceutical composition are also provided.
