An improved method for purifying albumin, a variant or fragment thereof, a fusion protein comprising albumin, a variant or fragment thereof, or a conjugate comprising albumin, a variant or fragment thereof is disclosed.

Method for purifying albumin

An affinity ligand-matrix conjugate of the structure Z-Spacer-[NX-A]-NY-A-NKis useful for the isolation, separation, purification, characterization, identification or quantification of endotoxins in an aqueous system, wherein m is an integer of at least one; each A independently represents an optionally substituted linear, branched or cyclic saturated hydrocarbon chain containing 1 to 6 carbon atoms; each X independently represents hydrogen or alkyl; Y is X or A-NX; and Z is a support matrix attached to the ligand through an optional spacer arm (Spacer).

Endotoxin-binding ligands and their use

The invention discloses methods for sequential protein isolation and purification from a biological sample by affinity chromatography. Affinity chromatography is conducted using ligands or ligand support complexes that selectively and specifically bind to proteins in the biological sample. The ligands or ligand support complexes were contacted sequentially in a predetermined order with the biological sample to allow each ligand or ligand-support complex to sequentially bind a protein from the biological sample.

Prion protein binding materials and methods for using the binding materials to detect or remove a prion protein from a sample, such as a biological fluid or an environmental sample. The binding materials are capable of binding to one or more forms of prion protein including cellular prion protein (PrPc), infectious prion protein (PrPsc), recombinant prion protein (PrPr), and proteinase resistant prion protein (PrPres). Prions from various species, including humans and hamsters, are bound by the binding materials.

Stable compositions containing biotinylated molecules, such as enzymes, are provided. The compositions include a biotinylated biomolecule, a biomolecule protectant, a buffer, a bulking agent selected from one or more water soluble, nonionic polymers and preferably a terminal sterilization protectant. The compositions can be utilized either as aqueous solutions or preferably in dried form, e.g., as a lyophilized, powder cake. They have applicability in any case where avidin/biotin technology is used and are particularly important as compositions containing a thrombin-like enzyme, e.g., for preparation of a fibrin monomer and fibrin monomer-based fibrin sealants.

A fibrin monomer/biotinylated enzyme mixture is formed. A material comprising an avidin/inert support composition having 1000 biotin binding units of activity per gram of the composition, a bulking agent and a protectant, is then introduced into the enzyme mixture to form a complex of the material and the biotinylated enzyme. The complex and enzyme are then removed therefrom to recover a fibrin monomer. Bulking agents selected from nonionic water soluble compounds are used.

A process is provided for the preparation of albumin which has extremely low levels of or is essentially free of colorants, metal ions, human proteins, fragments of albumin, polymers or aggregates of albumin, and viruses, and which is relatively non-glycated, relatively high in free thiol and with an intact C-terminus. The process comprises passing albumin (preferably expressed and secreted by transformed yeast) through two chromatography purifications, ultrafiltering the product, passing through two further chromatography steps and again ultrafiltering the product.
