The present invention relates to detection of nucleic acids and provides a composition comprising a Signal Generating Complex, wherein the composition comprises: (A) a pair of target probes (TPs), wherein a first TP of the pair of TPs comprises a nucleic acid sequence comprising two segments; (B) a pair of base PPAs comprising the first and second base PPAs, wherein the first base PPA comprises a nucleic acid sequence comprising three segments; (C) a set of extension PPAs comprising the first and second extension PPAs, wherein the first extension PPA comprises a nucleic acid sequence comprising two segments; (D) a plurality of pre-amplifiers (PAs), wherein the PAs comprise a nucleic acid sequence comprising three segments; (E) a plurality of amplifiers (AMPs), wherein the AMPs comprise a nucleic acid sequence comprising two segments; and (F) a plurality of label probes (LPs), wherein the LPs comprise a nucleic acid sequence comprising two segments.

Methods to further enhance signal amplification for the in situ detection of nucleic acids

The invention relates to methods of in situ detection of a nucleic acid variation of a target nucleic acid in a sample, including single nucleotide variations, multi-nucleotide variations or splice sites. The method can comprise the steps of contacting the sample with a probe that detects the nucleic acid variation or splice site and a neighbor probe; contacting the sample with pre-amplifiers that bind to the nucleic acid variation probe or splice site probe and neighbor probe, respectively; contacting the sample with a collaboration amplifier that binds to the pre-amplifiers; and contacting the sample with a label probe system, wherein hybridization of the components forms a signal generating complex (SGC) comprising a target nucleic acid with the nucleic acid variation or splice site, the probes and amplifiers; and detecting in situ signal from the SGC on the sample. The invention also provides samples, tissue slides, and kits relating to detection of nucleic acid variations, including single nucleotide variations, multi-nucleotide variations or splice sites, of a target nucleic acid.

In situ detection of nucleotide variants in high noise samples, and compositions and methods related thereto

A method of generating Source Identification information from a source packet stream and reliably transmitting the Source Identification information from a source to a destination over a communications channel is provided. The method operates on a set of source packets, wherein Source Identification information for each source packet to be transmitted is derived and delivered with the Source Identification information of all or most other source packets of an associated source block. The method includes techniques to minimize the network bandwidth required to deliver Source Identification information and techniques to overcome network impairments. When combined with FEC techniques, retransmission techniques, or combinations of FEC techniques and retransmission techniques, the methods described herein allow receivers to recover lost source packets, while simultaneously ensuring that the original source packets are not modified and thereby ensuring backwards compatibility for legacy receivers.

Generating and communicating source identification information to enable reliable communications

A sequence of symbol operations (a 'schedule representation') within a data storage device, wherein the operations are those used to process encoding or decoding operations of a forward error correction code (an 'FEC code') upon an arbitrary block of data of a given size (where size can be measured in numbers of symbols). The method is such that the schedule representation can be used to direct the processing of these operations upon a block of data in a way that is computationally efficient. Preferably, the same method can be applied to represent schedules derived from multiple different algorithms for the encoding or decoding of a code or for multiple different codes.

A method and apparatus for removing particles and residue that build up inside a substrate processing system during a substrate processing operation, without overetching system components, is described. One method includes the steps of: flowing an etchant gas comprising chlorine trifluoride (ClF.sub.3), diluted with an inert carrier gas, into a processing chamber after completion of the substrate processing operation. The parts of the system within the chamber with the greatest amount of build-up are preferentially heated to facilitate more extensive cleaning of those parts. Parts of the system within the chamber with less build up are protected from overetching by keeping them about 200.degree. C. cooler than the heavily-deposited parts. Heating the heavily-deposited chamber parts to a temperature of at least about 400.degree. C. allows using a lower concentration of etchant gas for the cleaning process than a lower temperature process would allow. The etchant gas reacts with both particles and residue in the chamber, reducing both particulate-related defects and deposition build-up. Another method includes blanketing lightly-deposited areas of the chamber with a nonreactive gas to displace and dilute the etchant gas from these areas for part of the cleaning process, while heavily-deposited areas are exposed to a higher concentration of the etchant gas for. a longer period of time. The blanketing gas is turned off for another part of the cleaning process so that these areas are also cleaned.
