The present invention related to methods and compositions for producing therapeutic proteins in yeast cell lines, and in particular, lacking dipeptidyl aminopeptidase (DAP) activity. DAP activity has been eliminated by genetically modifying acell line such that STE13 and DAP2 have been deleted.

Methods for increasing the yield and N-glycosylation site occupancy of paucimannose or complex N-glycans of recombinant glycoproteins produced in a recombinant host cell lacking dolichyl-P-Man:Man5GlcNAc2-PP-dolichyl alpha-1,3 mannosyltransferase (Alg3p) activity are disclosed. In particular, recombinant host cells are provided that comprise a disruption of the expression of an OS-9 family gene in the host cell. These recombinant host cells may then be used for producing recombinant glycoproteins. In further embodiments, the recombinant host cells further overexpress at least one heterologous single-subunit oligosaccharyltransferase, which in particular embodiments is capable of functionally suppressing the lethal phenotype of a mutation of at least one essential protein of the yeast oligosaccharyltransferase (OTase) complex.

Methods for increasing N-glycan occupancy and reducing production of hybrid N-glycans in pichia pastoris strains lacking ALG3 expression

Disclosed are methods for reducing detectable mannosylation of N-linked and O-linked oligosaccharides in lower eukaryote host cells. In particular, recombinant lower eukaryote host cells are provided in which expression of the GDP-mannose transporter encoded by the Vanadate Resistant Glycosylation 4 (VRG4) gene has been disrupted. In general, the VRG4 gene is essential for cell viability; however, the present invention provides host cells that are viable when expression of the VRG4 gene therein has been disrupted. The host cells are capable of producing proteins or glycoproteins that have reduced or no detectable &#x3b1;-linked mannose, &#x3b2;-linked mannose or phosphomannose containing N- and/or O-glycans.

Methods for reducing mannosyltransferase activity in lower eukaryotes

Methods for producing proteins and glycoproteins inthat lack detectable cross binding activity to antibodies made against host cell antigens are described. In particular, methods are described wherein recombinantstrains that do not display a &#x3b2;-mannosyltransferase 2 activity with respect to an N-glycan or O-glycan and do not display at least one activity selected from a &#x3b2;-mannosyltransferase 1, 3, and 4 activity to produce recombinant proteins and glycoproteins. These recombinantstrains can produce proteins and glycoproteins that lack detectable &#x3b1;-mannosidase resistant &#x3b2;-mannose residues thereon and thus, lack cross binding activity to antibodies against host cell antigens. Further described are methods for producing bi-sialylated human erythropoietin inthat lack detectable cross binding activity to antibodies against host cell antigens.

The present invention relates to eukaryotic host cells which have been modified to produce sialylated glycoproteins by the heterologous expression of a set of glycosyltransferases, including sialyltransferase and/or trans-sialidase, to become host-strains for the production of mammalian, e.g., human therapeutic glycoproteins. The present invention provides a novel lower eukaryotic host cell producing human-like glycoproteins characterized as having a terminal &#x3b2;-galactose residue and essentially lacking fucose and sialic acid residues. The present invention also provides a method for catalyzing the transfer of a galactose residue from UDP-galactose onto an acceptor substrate in a recombinant lower eukaryotic host cell, which can be used as a therapeutic glycoprotein.

A method for producing human-like glycoproteins by expressing a Class 2 &#x3b1;-mannosidase having a substrate specificity for Man&#x3b1;1,3 and Man&#x3b1;1,6 glycosidic linkages in a lower eukaryote is disclosed. Hydrolysis of these linkages on oligosaccharides produces substrates for further N-glycan processing in the secretory pathway.
