Disclosed are recombinant, synthetically and otherwise biologically produced novel proteins and polypeptides which are encoded by the DNA sequence for HSV-2 glycoprotein G (gG) or fragments of said gG sequence particularly by the unique sequence for gG or portions of said unique sequence. The unique sequence proteins and polypeptides are serologically active, can be produced easily and safely at low cost, are useful as diagnostic reagents for HSV-2 and as vaccines against HSV-2. Further disclosed are serological assays based on such unique sequence gG proteins and polypeptides that diagnose the presence of herpes simplex virus type 2 (HSV-2) specific antibodies and can differentiate between HSV-2 and herpes simplex virus type 1 (HSV-1) specific antibodies. Such assays are useful to diagnose genital infections, to detect for exposure to HSV-2 and to screen pregnant women to protect newborns from neonatal HSV-2 infection. Also disclosed are antibodies to such gG proteins and polypeptides which are useful therapeutically diagnostically and for affinity purification. Further, disclosed are purified and isolated DNA molecules which can be used as probes specific for HSV-2 DNA.

Herpes simplex virus type 2-glycoprotein G proteins and polypeptides

Disclosed are recombinant, synthetically and otherwise biologically produced novel proteins and polypeptides which are encoded by the DNA sequence for HSV-2 glycoprotein G (gG) or fragments of said gG sequence, particularly by the unique sequence for gG or portions of said unique sequence. DNA sequence for the unique sequence herpes simplex virus type

A canister for holding a solid phase support for use in a biospecific binding assay is disclosed. The canister consists of a receptacle and cap, both shaped to mate with each other in a snug fit, the cap having a central opening spanned by the solid phase in the form of a thin porous disk. The central opening is surrounded by a raised surface which forms a protective wall avoiding inadvertent contact of the disk by the user or surrounding objects. The cap and receptacle are constructed so as to provide a vent when the parts are joined, providing gravitational flow of a test sample across the porous disk into the interior of the receptacle.

Filter canister for immunoassays

A machine for transferring liquids to and from the wells of assay trays in a controlled, automated manner and a solid phase assay tray for use with the machine. The machine includes a horizontally translatable table (15) that holds the tray (46), a plurality of liquid dispensing manifolds (54) for dispensing liquids into the tray wells (50) and an aspirating manifold (65) for aspirating liquid from the well. The dispensing and aspirating manifolds are mounted on a vertically translatable head (16) above the table. Each dispensing manifold is equipped with a row of dispensing tubes (56) and is connected via a pump (58) to a liquid container (62). The aspirating manifold is equipped with a row of aspirating tubes (66) and is connected via a pump (69) to a waste liquid receptacle (73). A microprocessor (85) controls the movements of the table and manifolds and operates the pumps. The tray wells include means, such as sloping bottoms (74) or subwells (83), that cause the solid phase (76) to occupy a particular position in the wells and sumps (75), (84) that are positioned relative to the location of the solid phase such that they may be accessed vertically by the aspirating tubes without danger of disturbing the solid phase. The bottoms of the sumps have optically flat areas so that beams of light may be passed vertically through the liquid contents of the wells without intersecting the solid phase to make optical measurements of the liquid contents.

Automated assay machine and assay tray
