Disclosed herein are agents that target telomerase reverse transcriptase (TERT) for treating cancer and sensitizing cancer cells to genotoxic therapy. The present disclosure is directed to methods and/or uses of oligonucleotide conjugates for assays and flow cytometry detections and related systems and/or kits. Certain methods are directed to a method for detecting one or more biological targets of a sample in a detection assay, comprising: providing a molecular probe, comprising a binding moiety and an oligonucleotide sequence, to a sample comprising one or more biological targets; binding the one or more biological targets with the binding moiety; providing a detectable component to the sample, wherein the detectable component comprises a signal generating moiety conjugated to an oligonucleotide sequence complementary to the oligonucleotide sequence of the molecular probe; hydridizing the oligonucleotide sequence of the target-bound molecular probe to the detectable component; and detecting a signal generated from the hydridized detectable component. Various other embodiments, applications etc. are disclosed herein.

Methods and/or use of oligonucleotide conjugates for assays and flow cytometry detections

The instant disclosure provides reagent compounds, and antibody and oligonucleotide reagents, for use in a variety of assays, including immunoassays and nucleic acid hybridizations. The reagent compounds comprise a bridging antigen or bridging oligonucleotide and a latent crosslinker moiety, such as a tyramide moiety. The bridging antigens are recognizable by the antibody of a corresponding antibody reagent with high affinity, and the bridging oligonucleotides are complementary to the oligonucleotide of a corresponding oligonucleotide reagent. The antibody reagents and oligonucleotide reagents also comprise a crosslinker activation agent, such as a peroxidase enzyme. Reaction of the reagent compounds with the crosslinker activation agent results in the amplification of signal in assays for target cellular markers, including cellular antigens and nucleic acids. Also provided are detectable antibodies specific for the bridging antigens, kits comprising the reagent compounds and antibody and oligonucleotide reagents, methods of signal amplification using the compounds and reagents of the disclosure, methods of preparation of the compounds and reagents, and compositions comprising the compounds and reagents.

Reagent compounds, compositions, kits, and methods for amplified assays

Provided are apparatuses and methods for rapid viscometry of whole blood, plasma, and/or whole blood during coagulation. The disclosed technology measures the blood viscosity through the full range of flow rates found in the cardio-vascular system. This in vitro test can be performed on fresh or anticoagulated whole blood to predict the flow properties (e.g., viscosity) anywhere in the body from the aorta to the deep veins of the leg. The result is a flow-rate dependent blood viscosity curve (viscosity profile) that helps the clinician predict and manage the patient's vulnerability to thrombosis and embolism, which is of particular relevance to COVID-19 patients and/or ICU patients.

This invention is a novel method for analysis of data that is produced by test equipment. The preferred embodiment is data produced by liquid chromatography tandem mass spectrometry (LC-MS/MS) equipment, using industry standard methods to generate the initial data from the test equipment. The invention is a method for processing of the data to promptly produce accurate, reliable, and meaningful data that can be used for critical decisions. The unique benefit of the method is to correct the multiple measurement and calculation errors that are inherent in the operation of laboratory equipment. Prior methods result in errors based on circumstances that are difficult to control, accuracy-related errors in machine measurements, and fundamental mathematical errors in the data processing software that used with the laboratory equipment. As an added benefit, this novel method allows comprehensive simultaneous measurement and calculation of correlation of any and all peptide pairs in a single measurement, with the capability to support repeated measurements with changed conditions over time. This novel method allows robust, detailed, and comprehensive measurements of peptide activity and function, which results in substantial improvements over prior methods in accuracy, reliability, and efficiency.

Method for quantitative analysis of complex proteomic data

Methods and compositions for quantitative immunoassays are provided, in which ligand-conjugated probes are used to label samples and ligand-surfaced microspheres are used as quantitative reference standards. Certain embodiments provide a method of quantitative flow cytometry where ligands are oligonucleotides, and a sample comprising one or more cells is contacted with a hybridized antibody::fluorophore labeled targeting construct to label the cells, and the labeled cells are analyzed. In some embodiments, a population of quantitative labeled oligospheres labeled with the same fluorescent label as the cells is analyzed using the flow cytometer and used to create a quantitative standard curve of cytometer intensity versus molecules fluorescent label per oligosphere event. A standard curve trendline is established and used to determine the molecules of fluorescent label per cellular event for the antigen-positive cell populations. Based on molecules of fluorescent label per cell, the amount of Antibody Binding per Cell (ABC) is quantified.

The methods include generating reactions, and contacting each reaction generated with a set of internal standards, where each of the internal standards includes a different amount of product.
