The present invention relates to a method of treating obesity in a subject by administering to the subject a compound having the formula: wherein n is from 0 to 8; Ris CHOH, CH(OH)R, CHOR, COR, C(O)NH, C(O)NHR, C(O)NRR, C(O)NHNH, C(O)NHNHR, C(O)NHNRR, C(O)NRNH, C(O)NRNHR, C(O)NRNRR, C(O)NHNH(C(O)R), C(O)NHNR(C(O)R), C(O)NRNH(C(O)R), C(O)NRNR(C(O)R), CN, or C(O)R; Ris H, unsubstituted or substituted alkyl, YH, YR, YRR, YRYRYR, YC(O)R, C(O)YR, C(O)NH, C(O)NHR, C(O)NRR, NH, NHR, NRR, NHC(O)R, or NRC(O)R; Rand Rare the same or different and are selected from the group consisting of H, halogens, unsubstituted or substituted alkyl, OH, OR, NH, NHR, NRR, NHC(O)R, or NRC(O)R; R, R, R, R, R, R, and Rare the same or different and are selected from the group consisting of H, unsubstituted alkyl, substituted alkyl, unsubstituted aryl and substituted aryl; Ris selected from the group consisting of H, unsubstituted alkyl, and substituted alkyl; and Y is O or S; and pharmaceutically acceptable salts thereof.

Use of ibogamine congeners for treating obesity

A method for treating an addiction disorder (such as an addiction to or dependency on stimulants, nicotine, morphine, heroin, other opiates, amphetamines, cocaine, and/or alcohol) in a patient is disclosed. The method includes administering to the patient a first &agr; &bgr; nicotinic receptor antagonist and administering to the patient a second &agr; &bgr; nicotinic receptor antagonist. The second &agr; &bgr; nicotinic receptor antagonist is different than the first &agr; &bgr; nicotinic receptor antagonist, and the first &agr; &bgr; nicotinic receptor antagonist and the second &agr; &bgr; nicotinic receptor antagonist are administered simultaneously or non-simultaneously. Compositions which include a first &agr; &bgr; nicotinic receptor antagonist and a second &agr; &bgr; nicotinic receptor antagonist are also described. Examples of suitable &agr; &bgr; nicotinic receptor antagonists for use in the present invention's methods and compositions include mecamylamine, 18-methoxycoronaridine, bupropion, dextromethorphan, dextrorphan, and phamaceutically acceptable salts and solvates thereof. A method of evaluating a compound for its effectiveness in treating addiction disorders is also described.

The present invention is directed to compounds having formula (1), wherein n is from 0 to 8; R,is CH,OH, CH(OH)R,, CH,OR,, CO,R,, C(O)NH,, C(I)NHR,, C(O)NR,R,, C(O)NHNH,, C(O)NHNHR,, C(O)NHNR,R,, C(O)NR,NH,, C(O)NR,NHR,, C(O)NR,NR,R,, C(O)NHNH(C(O)R,), C(O)NHNR,(C(O)R,) C(O)NR,NH(C(O)R,), C(O)NR,NR,(C(O)R,), CN, or C(O)R,; R,is H, unsubstituted or substituted alkyl, YH, YR,, YC(O)R,, C(O)YR,, C(O)NH,, C(O)NHR,, C(O)NR,R,, NH,, NHR,, NR,R,, NHC(O)R,, or NR,C(O)R,; R,and R,are the same or different and are selected from the group consisting of H, halogens, unsubstituted or substituted alkyl, OH, OR,, NH,, NHR,, NR,R,, NHC(O)R,, or NR,C(O)R,; R,, R,, R,, R,, R,, R,, and R,are the same or different and are selected from the group consisting of unsubstituted alkyl and substituted alkyl and substituted alkyl; R,is selected from the group consisting of J, unsubstituted alkyl, and substituted alkyl; and Y is O or S; provided that when n is O, R,is selected from the group consisting of H, substituted alkyl, and unsubstituted alkyl; and pharmaceutically acceptable salts thereof. The compounds are useful in the treatment of subjects addicted to opiates and stimulants and have reduced side effects relative to other ibogamine congeners.
