The present invention provides a kit for preparing a composition for sealing a lung tract, comprising: (a) a first component comprising: (i) a fibrinogen solution, and (ii) prewet gelatin particles in an aqueous solution; and (b) a second component comprising: (i) a thrombin solution, and (ii) a dry gelatin powder; wherein the first and second components are stored separately and configured for mixing together to form a composition that is flowable and cross-linkable. The composition may be used to seal tissue tracts such as lung tissue tracts.

A method of sealing a resected surface of an organ includes applying a synthetic matrix to a resected surface of an organ, and applying an adhesive on the synthetic matrix so that the adhesive penetrates through interstices of the synthetic matrix for contacting an interface between the synthetic matrix and the resected surface of the organ. The method includes curing the adhesive for bonding the synthetic matrix to the resected surface of the organ. The synthetic matrix is a non-woven mesh made of polyglactin 910 or any other synthetic or non-synthetic fabric having a similar porosity or density. The adhesive is a biosynthetic or a synthetic adhesive. After penetrating through the pores of the synthetic matrix and curing, the cured biosynthetic or synthetic adhesive mechanically interlocks with the synthetic matrix for adhering the synthetic matrix to the tissue for creating a sealing barrier.

A composite implant is provided for repairing a tissue defect in a patient. In one embodiment, the implant is a porous tissue scaffold having at least one pocket formed therein and adapted to contain a viable tissue. The tissue scaffold can have a variety of configurations, and in one embodiment it includes top and bottom portions that can be at least partially mated to one another, and in an exemplary embodiment that are heated sealed to one another around a perimeter thereof to form an enclosed pocket therebetween. The pocket is preferably sealed with a viable tissue disposed therein. In another embodiment, the tissue scaffold is substantially wedge-shaped and the pocket comprises a hollow interior formed in the tissue scaffold, and/or at least one lumen extending into the tissue scaffold. The tissue scaffold can also optionally include at least one surface feature formed thereof to promote blood vessel formation.
