This application provides methods and compositions related to constructing nucleic acid hydrogels (e.g., RNA hydrogels) having repetitive monomer units, each monomer unit includes one or more G-quadruplex sequences. These G-quadruplex sequences cross-link the nucleic acid concatemer such that it self-assembles into a hydrogel under appropriate conditions. In some embodiments, each monomeric unit of the nucleic acid concatemer comprises a coding sequence for polypeptide of interest; and the nucleic acid hydrogel formed by the nucleic acid concatemer can be used for expressing the polypeptide in high quantities. In some embodiments, at least two RNA concatemers comprising G-quadruplex sequences are produced, one further comprising a spacer and the other further comprising a sequence encoding a polypeptide of interest. These two RNA concatemers are combined and self assembled to form a single, wideband RNA hydrogel.

A method of preparing a base sequence includes selecting candidate groups of base sequences of 5'-cohesive end having at least 50% GC content, calculating Gibb's free energy values of the selected candidate groups to re-select the candidate groups, and selecting the candidate groups, that have a sequence of 3 different successive bases among the base sequences from the re-selected candidate groups.

The present disclosure relates to a method of treating cancer using a composition including a target-specific self-illuminative nanocomplex which includes a self-illuminative nanocomplex including a quantum dot and a modified self-illuminative protein (m-Rluc8), a targeted cancer-specific molecule and a hydrophilic polymer, and a method of preparing the composition. Since the high efficiency self-illuminative nanocomplex may have high target specificity by chemically binding to a target-specific molecule, it can be effectively used in cancer treatment using a nanometer-scale laser light source, and therefore it is expected to propose a new paradigm of nanoptic therapeutics.

Method of treating cancer using high efficiency self-illuminative nanocomplexes and method of manufacturing the same

The present invention relates to a functional complex particle prepared by filling a nucleic acid hydrogel inside a liposome and a method of producing the same. The present invention may have an effect of increasing an expression of protein factors included in the particle by incorporating an X-shaped nucleic acid monomer in the nucleic acid hydrogel. Accordingly, when the core-shell particle is prepared using the method according to an embodiment of the present invention, an effect of facilitating an introduction of a genome into the nucleic acid hydrogel may be obtained, and thereby the core-shell particle may be used as a protein production platform copying a cell nucleus.

Core-shell nanoparticle including nucleic acid hydrogel and method of producing the same

The present invention relates to a multi-functional nucleic acid-based anti-cancer drug in which the anti-cancer drug is physically bound to a linear nucleic acid having a thiol group at 5' terminal thereof and then gold nanoparticles and aptamers are chemically bound. The present invention also relates to a method for preparing the anti-cancer drug and to an anti-cancer composition comprising the anti-cancer drug. The multi-functional nucleic acid-based anti-cancer drug according to the present invention uses A10 aptamer to achieve high targeting properties, and uses high-concentration anti-cancer drugs and gold nanoparticles to enable dual therapy of thermal therapy/chemical therapy, and may have less side effects and be more effective in anti-cancer therapy compared to existing anti-cancer drugs.
