Accurate measurements of the presence or absence of a target cell in a sample are provided. For example, the sample can be mixed with a plurality of transduction particles capable of binding to the target cells, the transduction particles being engineered to include a nucleic acid molecule formulated to cause the target cells to produce a plurality of detectable reporter molecules once the particles bind to and deliver the nucleic acid molecules into the one or more target cells. A set of signal data points are received that are associated with a quantity of reporter molecules and the signal data points are analyzed to accurately detect target cells in the sample. Systems and methods are disclosed.

Systems and methods for detecting cells using engineered transduction particles

Methods and systems are provided for packaging reporter nucleic acid molecules into non-replicative transduction particles for use as reporter molecules. The non-replicative transduction particles can be constructed from viruses and use viral transduction and replication systems. The reporter nucleic acid molecules include a reporter gene, such as a reporter molecule or selectable marker, for detecting target genes or cells. Methods and systems are provided for detection of cells and target nucleic acid molecules using the non-replicative transduction particles as reporter molecules.

Non-replicative transduction particles and transduction particle-based reporter systems

An apparatus includes a housing and an actuator. The housing, which defines a reagent volume that can receive a reagent container, can be removably coupled to a reaction chamber. The housing includes a puncturer that defines a transfer pathway in fluid communication with the reagent volume. A delivery portion of the housing defines a delivery pathway between the transfer pathway and the reaction chamber when the housing is coupled to the reaction chamber. The actuator has a plunger portion disposed within the reagent volume. An engagement portion of the actuator can be manipulated to move the plunger portion within the reagent volume to deform the reagent container. The puncturer can pierce a frangible portion of the reagent container to convey a reagent from the reagent container into the reaction chamber via the transfer pathway and/or the delivery pathway.
