The invention provides a method for the generation of a layered cellular 3 D microtissue aggregate, comprising the steps of contacting myeloid cells with a protein kinase C agonist, yielding primed myeloid cells; incubating the primed myeloid cells in the presence of LDL in a confined volume, particularly in a hanging drop culture; yielding a 3 D culture of myeloid cells; and incubating the 3 D culture together with fibroblasts in a hanging drop in the presence of LDL, yielding the layered cellular aggregate.

The invention relates to a method for preparing a polymer scaffold that comprises the steps of providing a piece of a fabric of filaments of a first biodegradable or biocompatible polymer, applying a coating of a second polymer to said arrangement of filaments, and stretching the piece along its axis of longitudinal extension, thereby obtaining an aligned microfibrillar scaffold. The invention further relates to a method for providing an artificial tissue, and to a microfibrillar scaffold of aligned filaments obtained by the method of the invention.

Expandable annulus sealing ring for stented minimally invasive heart valve prostheses

A biological heart valve replacement, particularly for pediatric patients, comprises a tubular segment (A) comprising a proximal end (Ep), a distal end (Ed) and a central portion (Pc) arranged between said proximal and distal ends and defining a longitudinal direction of the valve. The valve further comprises at least one inner leaflet (C) attached in hinge-like manner to a connection zone (F) at an inner wall (W) region of said central portion, each one of said inner leaflets being movable between a closing position and an opening position of the valve. In order to provide growth adaptability, the tubular segment comprises at least one tubular growth zone (B; B,B) configured as a longitudinal strip made of a growth-adaptive biomaterial, with the remainder of the tubular segment being made of a non-growth-adaptive biomaterial.

Biological heart valve replacement, particularly for pediatric patients, and manufacturing method

Various inserts, called shapers and spacers, are provided for controlling tissue engineered heart valve (TEHV) leaflet geometry during culture. These inserts will prevent TEHV leaflet retraction during culture, be able to control the leaflet geometry during culture, enable culturing TEHV leaflets with a larger coaptation area, control the height of the coaptation area, maintain TEHV leaflet curvatures, and/or enable possibilities to culture TEHV leaflets in open configuration.

Method for target and drug validation in inflammatory and/or cardiovascular diseases

Developing heart valves are exposed to dynamic strains by applying a dynamic pressure difference over the leaflets. The flow is kept to a minimum, serving only as a perfusion system, supplying the developing tissue with fresh nutrients. Standard heart valves were engineered based on B trileaflet scaffolds seeded with cells isolated from the human saphenous vein. Tissue compaction is constrained by the stent, inducing increasing pre-strain in the tissue. The dynamic strains the tissues are exposed to via the dynamic pressure difference, are estimated using finite element methods based on the mechanical properties of the neo-tissue, in order to get inside into the strain distribution over the leaflet.

Method of manufacturing a tissue-engineered prosthesis

In one aspect the invention provides a bioreactor system for the production of tissue prostheses. The system includes a bioreactor, a culture medium reservoir coupled to the bioreactor by a first conduit and a second conduit, a pump operable to pump fluid into and draw fluid out of a pumping chamber defined in the bioreactor to generate a pulsatile flow of culture medium through the reservoir and bioreactor via said first and second conduits, one or more flow meters operable to generate flow rate signals representative of the rate of culture medium flow through one or both of said first and second conduits, and a controller arranged to receive said flow rate signals from said one or more flow meters and to control the pump means in response to said received flow rate signals to provide a desired rate of culture medium flow.
