Synthesis of polyionenes with built-in degradable linkers through addition polymerization of a novel class of degradable A-type monomers (d-A), and their use as antimicrobial agents are disclosed. A library of biodegradable polyionenes and Gemini-surfactants made from d-Amonomers are also disclosed. These materials have potent and broad spectrum of antimicrobial activity with high selectivity over mammalian cells.

Biodegradable polyionenes

The present disclosure relates to polythioaminals with applications as carriers or delivery vehicles for therapeutic agents or other small molecule cargo. Polythioaminal block copolymer coupled to a therapeutic agent is a polymer-therapeutic conjugate that exhibits higher stability and longer life time in aqueous environments. The polythioaminal block copolymer coupled to a therapeutic agent can be synthesized by reacting hexahydrotriazines with a hydrophobic block precursor, a hydrophilic block precursor, a particle stabilizing segment precursor, and a cargo, such as a therapeutic agent, in a one pot synthesis. The ease of synthesizing the resulting polythioaminal block copolymer coupled to the therapeutic agent while offering the extended stability and polymer life time in aqueous environments make the polythioaminal block copolymer particularly attractive for therapeutic carriers.

Preparation of robust polythioaminal carriers

Eight-membered ring cyclic carbonates having an oxygen or an acylated nitrogen at position 6 were prepared by reaction of precursor diols with ethyl chloroformate. The cyclic carbonates undergo organocatalyzed ring opening polymerization. In one instance, the initial polymer formed comprises a carbonate repeat unit having a Boc-protected nitrogen in the polymer backbone. Deprotecting the nitrogen with acid forms a cationic carbonate repeat unit having a positive charged secondary ammonium nitrogen in the polymer backbone.

Cyclic carbonate monomers and ring opened polycarbonates therefrom

Cationic, anionic, and/or zwitterionic bis-urea compounds self-assemble by non-covalent interactions in aqueous solution to form high aspect ratio nanofibers. The nanofibers reversibly bind drugs by non-covalent interactions, forming drug compositions for exhibiting sustained release of the drug.

Self-assembling bis-urea compounds for drug delivery

An antimicrobial composition comprises an anionic drug and an amine polymer comprising a first repeat unit of formula (2): wherein a' is an integer equal to 1 or 2, b&#x2032; is an integer equal to 1 or 2, and each R&#x2032; is an independent monovalent radical selected from the group consisting of hydrogen, methyl, ethyl, and combinations thereof. G&#x2032; is a divalent linking group selected from the group consisting of a single bond and groups comprising at least one carbon. Xis a negatively charged counterion. The drug and the amine polymer are bound by noncovalent interactions.

Polycarbonates bearing pendant primary amines for medical applications

A cyclic carbonate monomer has the formula (2): wherein i) t and t' are integers independently having a value from 0 to 6 wherein t&#x2032; and t cannot both be zero, ii) each Qis a monovalent radical independently selected from the group consisting of hydrogen, halides, alkyl groups comprising 1 to 30 carbons, and aryl groups comprising 6 to 30 carbon atoms, iii) L&#x2032; is a divalent linking group comprising one or more carbons, and iv) S&#x2032; is a steroidal group.

Polycarbonates for delivery of drugs and methods of preparation thereof

A composite hydrogel comprises an amphiphilic triblock copolymer (ABA) and a loaded micelle bound by noncovalent interactions. The loaded micelle comprises a biologically active substance and an amphiphilic diblock copolymer (CD). The A blocks comprise a steroidal repeat unit (repeat unit 1) having both a backbone carbonate and a side chain bearing a steroid functional group. Each of the A blocks has a degree of polymerization of about 0.5 to about 4.0. The B block comprises a first poly(alkylene oxide) backbone. The C block comprises a second poly(alkylene oxide) backbone. The D block comprises a steroidal repeat unit (repeat unit 2) having both a backbone carbonate group and a side chain comprising a steroid functional group. The composite hydrogel is capable of controlled release of the biologically active substance.
