The present disclosure provides methods for detecting the presence of a cancer stem cell and their use in cancer prognosis, evaluating risk of cancer metastasis, identifying or validating drug candidates, and determining treatment efficacy. It also provides kits useful for detecting the presence of cancer stem cells as well as methods of treating cancer using CAIX inhibitors.

Carbonic anhydrase IX-related markers and use thereof

Therapeutic ureido-sulfonamide compositions having compounds with the formulaR-Q-Ar&#x2014;SONHare disclosed, which compounds selectively inhibit CAIX and CAXII, and which are effective to inhibit hypoxic tumor growth, suppress metastases, and impair and deplete cancer stem cells in mammals.

Sulfonamide compounds for inhibition of metastatic tumor growth

There is provided a method for determining tumour metastatic potential using biomarkers of tumour metastasis comprising any two of carbonic anhydrase-9 (CAIX), vascular endothelial growth factor C (VEGF-C), ephrin A5 (EFNA5), eph receptor B2 (EPHB2), transforming growth factor beta 3 (TGF-&#x3b2;3), pyruvate dehydrogenase kinase isoenzyme-3 (PDK3), carbonic anhydrase-12 (CAXII), keratin 14 (KRT14), hypoxia inducible factor 1 alpha subunit (HIF-1&#x3b1;), and tenascin C (TNC). CAIX, VEGF-C, EFNA5, EPHB2, TGF-&#x3b2;3 or PDK3 may be indicators of moderate metastatic potential, while CAXII, KRT14, HIF-1&#x3b1;, or TNC may be indicators of high metastatic potential. The biomarkers may be used to assess malignancies or cancers having hypoxic regions, such as breast cancer.

Biomarkers of cancer metastasis

Inhibitors of integrin-linked kinase (ILK) are used in the treatment of inflammatory disease, including cutaneous inflammatory diseases, such as psoriasis, scleroderma, systemic lupus erythematosus and atopic dermatitis.

ILK genetic sequences and methods of use are provided. In some embodiments, a fragment or portion of an ILK polynucleotide is provided, where the polynucleotide has a sequence consisting of contiguous nucleotides within the provided ILK sequences, which may be within the provided ILK coding sequences. Such polynucleotides may be DNA, RNA, and the like, and may be double stranded or single stranded.

The ILK nucleic acid compositions find use in identifying homologous or related proteins and the DNA sequences encoding such proteins; in producing compositions that modulate the expression or function of the protein; and in studying associated physiological pathways. In addition, modulation of the gene activity in vivo is used for prophylactic and therapeutic purposes, such as identification of cell type based on expression, and the like.
