A method for improving intestinal flora increases the number of beneficial bacteria such as lactic acid bacteria andto improve intestinal flora by using an enzyme. The method involves providing a protease, which is a polypeptide having an amino acid sequence shown in SEQ ID NO:that can increase beneficial bacteria such as lactic acid bacteria andin intestines to exert an excellent effect of improving intestinal flora.

Method for improving intestinal flora

The purpose of the present invention is to provide an agent for improving intestinal flora which can increase the number of beneficial bacteria such as lactic acid bacteria andto improve intestinal flora by using an enzyme. A protease consisting of a polypeptide having an amino acid sequence shown in SEQ ID NO: 1 can increase beneficial bacteria such as lactic acid bacteria andin intestines to exert an excellent effect of improving intestinal flora.

Intestinal flora improvement agent

The purpose of the present invention is to provide an agent for improving intestinal flora which can increase the number of beneficial bacteria such as lactic acid bacteria andto improve intestinal flora by using an enzyme. An amylase that is derived from a microorganism belonging tocan increase beneficial bacteria such as lactic acid bacteria andin intestines to exert an excellent effect of improving intestinal flora.

The present invention has a purpose of providing a technique for increasing the heat resistance of a &#x3b2;-galactosidase. According to the present invention, in a reference &#x3b2;-galactosidase amino acid sequence which shows a 90% or more identity to the amino acid sequence of SEQ ID NO: 4, proline is substituted for one or more amino acids selected from the group consisting of the following amino acids: (1) an amino acid corresponding to lysine at position 166 of the amino acid sequence of SEQ ID NO: 4, (2) an amino acid corresponding to glycine at position 307 of the amino acid sequence of SEQ ID NO: 4, and (3) an amino acid corresponding to alanine at position 833 of the amino acid sequence of SEQ ID NO: 4.

A method for making a mutated pullulanase enzyme that hydrolyzes an &#x3b1;-1,6-glycosidic linkage is provided. The method includes obtaining the amino acid sequence of a pullulanase enzyme having an amino acid sequence that is at least 95% identical to the amino acid sequence set forth in SEQ ID NO: 13; identifying an amino acid to be mutated in the pullulanase enzyme of step (1), wherein the amino acid to be mutated corresponds to the amino acid at position Phe476 of SEQ ID NO: 2; constructing a mutated amino acid sequence by substituting the amino acid to be mutated with another amino acid or deleting the amino acid to be mutated, thereby making a mutated pullulanase enzyme having the mutated amino acid sequence that has increased affinity for pullulan and that hydrolyzes an &#x3b1;-1,6-glycosidic linkage.
