Markers for the diagnosis of biochemical recurrence in prostate cancer

The present disclosure provides gene expression profiles that are associated with prostate cancer, including certain gene expression profiles that differentiate between subjects of African and Caucasian descent and other gene expression profiles that are common to subjects of both African and Caucasian descent. The gene expression profiles can be measured at the nucleic acid or protein level and used to stratify prostate cancer based on ethnicity. The gene expression profiles can also be used to identify a subject for prostate cancer treatment. Azophenols as ERG oncogene inhibitors

Monoclonal antibodies, or antigen-binding fragments thereof, that bind to ERG, and more specifically, to an epitope formed by amino acids 42-66 of ERG3 are disclosed. The monoclonal antibodies can be non-human antibodies (e.g., rabbit or mouse) or humanized monoclonal antibodies having the CDR regions derived from those non-human antibodies. In other embodiments, the monoclonal antibodies are chimeric, having the light and heavy chain variable regions of a non-human ERG antibody. Methods of using the antibodies to detect ERG, or fusion proteins comprising all or part of an ERG polypeptide, such as an ERG polypeptide encoded by a TMPRSS2/ERG, SLC45A3/ERG, or NDRG1/ERG fusion transcript, are also provided, including methods of detecting ERG or ERG fusion events in a clinical setting. The antibodies can also be used to inhibit the activity of ERG or fusion proteins comprising all or part of an ERG polypeptide, such as an ERG polypeptide encoded by a TMPRSS2/ERG, SLC45A3/ERG, or NDRG1/ERG fusion transcript and to treat malignancies associated with overexpression of ERG or an ERG fusion event, such as prostate cancer, Ewing's sarcoma, acute myeloid leukemia, acute T-lymphoblastic leukemia, endothelial cancer, and colon cancer.

Provided are therapeutic compositions and methods of detecting and treating cancer, including prostate and other related cancers. The present disclosure provides genomic arrangements of the chromosome 3q13 region that are associated with prostate cancer, such as rearrangements between the ZBTB20 and LSAMP genes, including gene fusions between the ZBTB20 gene and the LSAMP gene and deletions spanning both genes. The ZBTB20/LSAMP genomic rearrangement serves as a biomarker for prostate cancer and can be used to stratify prostate cancer based on ethnicity or the severity or aggressiveness of prostate cancer and/or identify a patient for prostate cancer treatment. Another aspect involves discovering that deletions of the PTEN gene are observed predominately in prostate cancer from subjects of Caucasian descent. The gene expression profiles can be used to detect prostate cancer cells in a sample and to distinguish between well differentiated (WD) prostate cancer and poorly differentiated (PD) prostate cancer. Alterations to the ERG gene are some of the most prominent genomic changes associated with many cancers including prostate cancer. Such cancer specific alterations result in the overexpression of wild type ERG protein or an altered ERG protein. The present invention provides selective inhibitors of wild type or an altered ERG protein expression. The ERG inhibitors of the invention, therefore, are therapeutic agents for treating ERG positive cancers.
