Disclosed is a flow imaging cytometer having a flow cell one side of which is irradiated with excitation light, in which wavelength selecting means for reflecting excitation light and transmitting fluorescence or reflecting means for reflecting excitation light is provided on the side of the flow cell opposite the side irradiated, thereby increasing the intensity of the excitation light so that the intensity of fluorescence emitted from an irradiated cell is increased. It is possible to pick up the fluorescent images at all times. The cytometer includes a light source for emitting the excitation light, dichroic mirrors which construct the wavelength selecting means, a photomultiplier for detecting fluorescence from irradiated cells, and a cell flow-by decision circuit which, based upon a detection signal from the photomultiplier, determines whether a detected cell of interest is suited to pick-up of a fluorescent image and produces a prescribed control signal for controlling an electronic shutter.

An apparatus for forming a flattened sample flow by passing a sample (liquid specimen) containing particle components such as blood and urine, in a broad, thin, flat flow. This apparatus is preferably used in an apparatus for analyzing particle images by emitting a strobe light to a sample flat flow, and taking the still images of the particle components. More specifically, the cross section of a measuring passage of a flow cell is rectangular with a side ratio of one to several times, with a shape that gradually narrows in width only in one direction of the lead-in passage, and the discharge port at the front end of the sample nozzle has a flat opening, or small discharge ports are arranged horizontally in a row. The decreasing direction of the width in the lead-in passage and the shorter diameter direction of the flat discharge port or the direction of the small discharge ports arranged horizontally in a row are matched. Besides, the cross section of the measuring passage of the flow cell is rectangular with a side ratio of one to several times, and sheath liquid dividing means disposed at the discharge port upstream side of the sample nozzle for dividing the sheath liquid into two symmetrical flows. The sample nozzle is placed so that the discharge port may be positioned at the converging region of the sheath liquid. For further enhancing the flatness of the sample flow, a sample nozzle having a flat discharge port, or plural discharge ports disposed horizontally in a row is used.

Apparatus for forming flattened sample flow for analyzing particles

A flow imaging cytometer, in which an image capturing area for cells traveling in the flow of a specimen solution is constantly monitored to perform cell photography in an efficient manner, is improved so that fluorescent images of cells can be acquired with greater efficiency. Cell particles to be sensed in the specimen are treated with a fluorescent stain, and the specimen solution is irradiated with light from a pulsed light source for exciting fluorescence, and with infrared light constantly for monitoring the passage of cells through the cytometer. When the monitoring light is made to serve also as the light for exciting fluorescence, the specimen flow is continuously irradiated at all times at a low luminance for the purpose of monitoring. Then, when a cell particle of interest is detected, the cell particle is irradiated with a light pulse of high luminance to obtain a still picture of the fluorescence-emitting cell. At detection of the cell, a source of white light can be actuated to acquire a white-light image of the cell.

A flow imaging cytometer has an image capturing area for capturing the white-light image of a cell, and an image capturing area for capturing a fluorescent image of the same cell. The two image capturing areas are positionally offset from each other so that both images can be obtained in an excellent state with little mutual influence from the two types of light. Based upon detection of a cell using light from a light source for monitoring cell flow-through, a light source for exciting fluorescence and a strobe light source for emitting the white light area actuated after respective prescribed time delays. To this end, the cytometer incorporates a delay-pulse generating circuit serving as control means.

An imaging flow cytometer is provided with a continuous-emission light source for continuously monitoring cells passing through the image capturing area of a video camera for cell-image capturing, and with an excitation light source for picking up a fluorescent image of a cell. When a line sensor monitoring cell passage through the cytometer senses such cell passage, the cell is irradiated with strobe light and then, after waiting for the cell to move a fixed distance, with the excitation light. Thus, a particle analyzer is provided in which an image by white light resulting from the strobe light and a fluorescent image resulting from the excitation light can be captured simultaneously by a single video camera in either upper and lower halves or right and left halves of one imaged frame.
