A method for the preparation and purification of compounds using a novel support, a tetrabenzo &lsqb;a, c, g, i&rsqb;-fluorene group (Tbf) comprising reacting a building block (A) containing a Tbf group (Tbf-A), with a second building block (B), to afford an intermediate compound (Tbg-A-B) followed by purifying the intermediate compound by adsorption on a carbon support, removing the intermediate compound from the support with a solvent and repeating the previous reactions using the required number of building blocks to synthesize the compounds followed by removal of the Tbf group to afford the desired compounds.

Support for synthesis and purification of compounds

Multiple fluid sample processors and systems for high throughput chemical synthesis and biological assays and/or processing. A multi-layered fluidic array having microchannels, reservoirs and reaction wells is subject to robotic and automated handling. A pressure pumping system is utilized for fluid delivery and control through the synthesis process. The sizes of the micro-sized channels, apertures, and valves are adjusted to optimize fluid distribution and channel filling. The fluid sample processors can be grouped together in a microtiter format to increase the speed, quantity and efficiency of the processing.

Multiple fluid sample processor and system

An apparatus and method for phase separation of two materials is disclosed. The two materials to be separated are positioned in an extraction cartridge and passed through a filter device, the filter device includes a filter disk member positioned adjacent at least one and preferably two frit members. A drying cartridge may be used to dry the separated liquid material after it leaves the extraction cartridge. The invention can be used for reaction workup, quenching, product isolation and purification.

Methods and apparatus for liquid phase separation

A wash station for washing probes which are used with robotic systems. In particular, the wash station is used with robotic systems to provide simultaneous cleaning of both the interior and the exterior of multiple probes employed by the system. The wash station consists of a base, multiple chambers and multiple cylinders. The robotic probe to be cleaned is inserted within one of the cylinders where it is immersed in wash liquid to wash the exterior walls. The cylinders are separated by chambers. The wash liquid is removed from the wash station via a drain located in the base. In a preferred embodiment, independent access for each probe into the wash station is provided. In addition, the wash station provides a large capacity for fluid handling and is constructed such that the station is water tight. In a particularly preferred embodiment, the wash station is constructed from chemically compatible materials which will not be affected by exposure to the materials needed for general chemical synthesis.

The apparatus consists of: a reservoir block having a plurality of wells; a plurality of reaction tubes, usually gas dispersion tubes, having filters on their lower ends; a holder block, having a plurality of apertures; and a manifold, which may have ports to allow introduction/maintenance of a controlled environment. The manifold top wall has apertures and a detachable plate with identical apertures. The apparatus is constructed from materials which will accommodate heating, cooling, agitation, or corrosive reagents. Gaskets are placed between the components. Rods or clamps are provided for fastening the components together. Apparatus operation involves placing the filters on the lower ends of the reaction tubes in the reservoir block wells, and the upper ends passing through the holder block apertures and into the manifold. The apparatus provides in excess of 1 mg of each product with structural knowledge and control over each compound. The apparatus can be adapted to manual, semiautomatic or fully automatic performance. Using the apparatus a series of building blocks are covalently attached to a solid support. These building blocks are then modified by covalently adding additional different building blocks or chemically modifying some existing functionality until the penultimate structure is achieved. This is then cleaved from the solid support by another chemical reaction into the solution within the well yielding an array of newly synthesized individual compounds, which after postreaction modification, if necessary, are suitable for testing for activity.

Apparatus and method for multiple simultaneous synthesis

A system for the multiple, simultaneous synthesis of compounds preferably uses a reaction apparatus comprising a reservoir rack having a plurality of reaction wells, a plurality of reaction tubes, usually gas dispersion tubes, having filters on their lower ends, a holder block, having a plurality of apertures, and a manifold, which has ports to allow introduction/maintenance of a controlled environment. The manifold top wall has a plurality of apertures in axial alignment with the reaction tubes and a gasket which allows penetration by a needle in order to dispense and aspirate materials from the reaction tubes. Sealing members, such as gaskets, are placed between the holder block, manifold and reservoir rack and the components are releasably fastened together. A robotic sample processor is used to automate the synthesis process using the reaction apparatus.
