Methods and devices using non-autologous mesenchymal stem cells comprising treating a recipient in need thereof with an effective amount of non-autologous mesenchymal stem cells, including methods and preparations for treating and regenerating connective tissue and enhancing bone marrow engraftment in an individual; and for using genetically engineered allogeneic human mesenchymal stem cells that carry within them genes of interest particularly for the expression of physiologically or pharmacologically active proteins in gene therapy for correction of genetic disorders or 'rebuilding' proteins important in tissue repair.

Uses for non-autologous mesenchymal stem cells

Disclosed are compositions and methods for augmenting bone formation by administering isolated human mesenchymal stem cells (hMSCs) with a ceramic material or matrix or by administering hMSCs; fresh, whole marrow; or combinations thereof in a resorbable biopolymer which supports their differentiation into the osteogenic lineage. Contemplated is the delivery of (i) isolated, culture-expanded, human mesenchymal stem cells; (ii) freshly aspirated bone marrow; or (iii) their combination in a carrier material or matrix.

Biological material for the repair of connective tissue defects comprising mesenchymal stem cells and hyaluronic acid derivative

Disclosed are compositions and methods for accelerating the differentiation of human MSCs into the osteogenic lineage with resultant enhanced de novo bone formation. This makes possible improved methods for bone defect repair. Thus, one aspect of the invention is a method for accelerating the differentiation of ALCAM-bearing hMSCs into the osteogenic lineage by contacting such hMSCs with a ligand that binds to activated leukocyte-cell adhesion molecule (ALCAM) on a single hMSC. Preferably, the ligand is a F fragment of a monoclonal antibody expressed by the hybridoma of ATCC Accession No. NB 11789.

Ligands that modulate differentiation of mesenchymal stem cells

Methods and devices using non-autologous mesenchymal stem cells comprising treating a recipient in need thereof with an effective amount of non-autologous mesenchymal stem cells, including methods and preparations for treating and regenerating connective tissue and enhancing bone marrow engraftment in an individual; and for using genetically engineered allogeneic human mesenchymal stem cells that carry within them genes of interest particularly for the expression of physiologically or pharmacologically active proteins in gene therapy for correction of genetic disorders or &ldquo;rebuilding&rdquo; proteins important in tissue repair.

Disclosed are compositions and methods for accelerating the differentiation of human MSCs into the osteogenic lineage with resultant enhanced de novo bone formation. This makes possible improved methods for bone defect repair. Thus, one aspect of the invention is a method for accelerating the differentiation of ALCAM-bearing hMSCs into the osteogenic lineage by contacting such hMSCs with a ligand that binds to activated leukocyte-cell adhesion molecule (ALCAM) on a single hMSC. Preferably, the ligand is a F.sub.ab fragment of a monoclonal antibody expressed by the hybridoma of ATCC Accession No. HB 11789.

Methods for in vitro or ex vivo lineage directed induction of isolated, culture expanded human mesenchymal stem cells comprising contacting the mesenchymal stem cells with a bioactive factor effective to induce differentiation thereof into a lineage of choice as well as such compositions including isolated culture expanded human mesenchymal stem cells and bioactive factors effective to induce directed lineage induction are disclosed. Further disclosed is this method which also includes introducing such culturally expanded lineage-induced mesenchymal stem cells into a host from which they have originated for purposes of mesenchymal tissue regeneration or repair.

Lineage-directed induction of human mesenchymal stem cell differentiation

Monoclonal antibodies specific for normal human osteogenic cell subsets and the monoclonal hybridoma cell lines that synthesize and secrete these monospecific antibodies, as well as the uses of the monoclonal antibodies for diagnostic and/or therapeutic purposes.
