A method of sample preparation for streamlined multi-step assays is provided. The method includes the step of providing a microfluidic device including a reservoir defined by a surface configured to repel an aqueous solution. A dried reagent is provided on a portion of the surface and the reservoir is filled with an oil. A first droplet formed from the aqueous solution is positioned on the dried reagent so to pick-up and re-dissolve the dried reagent therein so as to expose the portion of the surface. In addition, a second droplet of an aqueous solution may be deposited on a hydrophilic spot patterned on the surface. A magnetic force may be configured to interact magnetically with the paramagnetic beads within the first droplet to move the droplet through the oil in the reservoir or to move the paramagnetic beads from the first droplet, through the oil, into the second droplet.

Volume-free reagent addition and exclusion-based sample preparation for streamlined multi-step assays

A device and a method for isolating a target from a sample are provided. The target is bound to solid phase substrate to form a target bound solid phase substrate. The device includes a first plate having a first region for receiving at least a portion of the sample. A second plate is spaced from the first plate by a distance and has a first region for receiving a reagent. A force attracts the target bound solid phase substrate toward the first region of the second plate such that the target bound solid phase substrate in the portion of the sample are drawn through the air gap and into the reagent by the force.

Device and method for extracting a targeted fraction from a sample

A device and a method for isolating a target from a biological sample are provided. The target is bound to solid phase substrate to form target bound solid phase substrate. The device includes a lower plate with an upper surface having a plurality of regions. The biological sample is receivable on a first of the regions. An upper plate has a lower surface directed to the upper surface of the lower plate. A force is positioned adjacent the upper plate and attracts the target bound solid phase substrate toward the lower surface of the upper plate. At least one of the upper plate and the lower plate is movable from a first position wherein the target bound solid phase substrate in the biological sample are drawn to the lower surface of the upper plate and a second position wherein the target bound solid phase substrate are isolated from the biological sample.

A method is provided for facilitating extraction of a fraction from a biological sample. The biological sample includes non-desired material and a fraction-bound solid phase substrate. The method includes the steps of capturing the fraction-bound solid phase substrate and bringing an isolation buffer and the fraction-bound solid phase substrate into contact to purify the captured fraction-bound solid phase substrate.

Method for facilitating extraction of a fraction from a biological sample

A device and a method are provided for isolating a fraction in a biological sample. The fraction is bound to solid phase substrate to define a fraction-bound solid phase substrate. The device includes an input zone for receiving the biological sample therein to capture a desired fraction of the biological sample. A force is provided that is generally perpendicular to gravity. The force is movable between a first position adjacent the input zone multiple other positions adjacent various purification, protein analysis, separation and extraction zones. The force captures the fraction-bound solid phase substrate and the fraction-bound solid phase substrate moves from the input zone to the other zones to perform a multi-step assay on the isolated fraction within the device.

Device for and method of isolating and analyzing a fraction in a biological sample

A device and method are proved for transferring a target from a first location to a second location. The target is bound to solid phase substrate to form a target bound solid phase substrate. The device includes transfer surface for receiving the target bound solid phase substrate thereon for transfer. The transfer surface movable between a first position wherein the transfer surface is aligned with the first location and spaced therefrom by a distance and a second position wherein the transfer surface is aligned with the second location. An alignment structure aligns the transfer surface with respect to the second location, with the transfer surface in the second position. A force is movable between an attraction position wherein the target bound solid phase substrate are drawn toward the transfer surface and a discharge position wherein the target bound solid phase substrate are free of the force.

Method for transferring a target between locations

A method and microfluidic device are provided for containing a droplet having an outer surface at a predetermined location. The microfluidic device includes a plate having an upper surface and a central region communicating with the upper surface. The central region is adapted for receiving a droplet of fluid thereon. The central region includes an outer periphery that defines a first fluid constraint configured for discouraging fluid on the central region from flowing therepast. A second fluid constraint extends about the first fluid constraint. The second fluid constraint is configured for discouraging fluid flowing therepast. A third fluid constraint extends about the second fluid constraint. The third fluid constraint configured for discouraging fluid flowing therepast.

Method and device for containing expanding droplets

A deformable well structure for a microtiter plate and method are provided. The deformable well structure includes a sample container defining a well for receiving a sample therein. The sample received in the well has a concave meniscus. A deformation tool is engageable with the sample container and is moveable between a first disengaged position wherein the deformation tool is spaced from the sample container and a second, engaged position wherein the deformation tool engages and deforms at least a portion of the sample container such that the meniscus of the sample in the well is converted from concave to convex.

An adapter for a sliding head of a sample processing system is provided that includes a plate and a magnet mounting recess wall. The plate includes a top surface, a bottom surface, a front wall extending between the top surface and the bottom surface, and a back wall extending between the top surface and the bottom surface. The magnet mounting recess wall is mounted to the top surface. The magnet mounting recess wall is configured to accommodate a magnet of a sliding head of a sample processing system. At least a portion of the bottom surface is curved in a first direction from the front wall to the back wall and is concave relative to the top surface.

A sample plate includes a top surface, a plurality of wells, and a plurality of reservoirs. The plurality of wells is mounted to the top surface. Each well includes a well bottom surface and a well wall extending up from the well bottom surface. The plurality of reservoirs is mounted to the top surface. Each reservoir includes a reservoir bottom surface, a reservoir wall extending from a first side of the reservoir bottom surface, and the well wall extending from a second side of the reservoir bottom surface. Each reservoir of the plurality of reservoirs surrounds a corresponding well of the plurality of wells.

A device and a method are provided for isolating a fraction in a biological sample. The fraction is bound to solid phase substrate to define a fraction-bound solid phase substrate. The device includes an input zone for receiving the biological sample therein and a second zone for receiving an isolation fluid therein. A force is provided that is generally perpendicular to gravity. The force is movable between a first position adjacent the input zone and a second position adjacent the isolation zone. The force captures the fraction-bound solid phase substrate and the fraction-bound solid phase substrate moves from the input zone to the isolation zone in response to the force moving from the first position to the second position.

Device for isolating a fraction in a biological sample

Methods of isolating weakly interacting molecules in a fluidic sample using an immiscible phase filtration technique are disclosed. A complex is formed between a solid phase substrate, a molecule immobilized on the solid phase substrate, and at least one target molecule present in the fluidic sample. The complex is transferred into an immiscible phase by applying an external force to the solid phase substrate. The methods eliminate the need for complex and time consuming washing steps.
