The invention is directed to products and methods for preparing self-seeding vascular constructs generated as a bi-layered electrospun matrices, conjugated with EPC-specific antibodies and anti-thrombogenic agents on the inner surfaces of their lumens.

Vascular constructs

Provided are live, artificial, skin substitute products and methods of making and using the same, such as for wound treatment and compound testing, including compound testing for efficacy, toxicity, penetration, irritation and/or metabolism testing of drug candidates or compositions such as cosmetics.

Multi-layer skin substitute products and methods of making and using the same

A method of making an organ or tissue comprises: (a) providing a first dispenser containing a structural support polymer and a second dispenser containing a live cell-containing composition; (b) depositing a layer on said support from said first and second dispenser, said layer comprising a structural support polymer and said cell-containing composition; and then (c) iteratively repeating said depositing step a plurality of times to form a plurality of layers one on another, with separate and discrete regions in each of said layers comprising one or the other of said support polymer or said cell-containing composition, to thereby produce provide a composite three dimensional structure containing both structural support regions and cell-containing regions. Apparatus for carrying out the method and composite products produced by the method are also described.

Integrated organ and tissue printing methods, system and apparatus

The invention is directed to methods of inducing cell recruitment and tissue regeneration at a target site in a subject. It is also based, in part, on the discovery that a subject's own biologic resources and environmental conditions can be used for in situ tissue regeneration and thereby reduce or eliminate the need for donor cell procurement and ex vivo manipulation of such donor cells. Methods are disclosed for recruitment of a subject's own stem cells to a target region by inducing a sustained positive pressure at a target site, such as the kidney, thereby increasing the number of pluripotent cells capable of differentiating to regenerate the target tissue.

Directed stem cell recruitment

Vascular scaffolds and methods of fabricating the same are disclosed for tissue engineering of vascular constructs. By combining electrospun matrices with cell sheet technologies, vascular constructs with more mature cell layers can be obtained for reconstruction of blood vessels, heart valves and the like. A engineered smooth muscle cell sheet, wrapped around an electrospun vascular scaffold, is able to provide a mature SMC layer that expresses strong cell-to-cell junction markers and contractile proteins. In addition, preconditioning of the cell sheet covered vascular scaffold maintained cell viability and infiltration into the scaffold.

Laminous vascular constructs combining cell sheet engineering and electrospinning technologies

Provided are live, artificial, skin substitute products and methods of making and using the same, such as for wound treatment and compound testing, including compound testing for efficacy, toxicity, penetration, irritation and/or metabolism testing of drug candidates or compositions such as cosmetics. Described herein is an artificial mammalian skin substitute product, comprising: (a) optionally, but in some embodiments preferably, a first ('hypodermis-like') layer comprising live mammalian adipocytes (e.g., induced pre-adipocytes) in a first hydrogel carrier; (b) a second ('dermis-like') layer contacting or directly contacting the first layer and comprising live mammalian fibroblast cells and' live mammalian follicle dermal papilla cells in combination in a second hydrogel carrier; (c) a third (&#x201c;epidermis-like&#x201d;) layer contacting or directly contacting the second layer (i.e., on the opposite side thereof as the first layer, so that the second layer is sandwiched between the first and third layers when the first layer is present), the third layer comprising live mammalian keratinocytes and live mammalian melanocytes in combination in a third hydrogel carrier.

Provided herein is a tissue stretching device configured for orbicular expansion of a tissue placed therein. Methods of use of the device to stretch a tissue as well as for culturing organized tissues are also provided. Stretched and/or cultured tissues produced by these processes are also provided, as well as methods making use of the same.

Provided herein is a tissue stretching device including a tissue clamping member defining an area in a Z plane, wherein the tissue clamping member is configured to hold tissue parallel to the Z plane. Methods of use of the tissue stretching device to stretch a tissue as well as for culturing organized tissues are also provided. Stretched and/or cultured tissues produced by these processes are also provided, as well as methods of treatment making use of the same.

Tissue expander

Provided herein are anisotropic muscle implants that have a biodegradable scaffold comprising a plurality of fibers oriented along a longitudinal axis. The implants may include mammalian muscle cells seeded and/or fused into myotubes on the scaffold. Methods of forming the muscle implants are provided, as are methods of treating a subject in need of skeletal muscle reconstruction.
