Spheroid microtissues that can mimic native tissue-like structure and function, spheroid production methods that are high-throughput, suitable for efficient production, maintainable over long-term culture, and/or offer repeatable control over size distribution. Spheroids that have blood vessels, including spheroids with functional, blood-perfused vascular networks upon injection in vivo. Dissolvable hydrogel microwell arrays for high throughput parallel formation of spheroids in a single pipetting step and easy retrieval for downstream applications. A method to produce prevascularized microtissues in sufficient numbers to form a macrotissue in vivo for therapeutic purposes. This method is based on sacrificial release of dissolvable microwell templates, a novel and scalable strategy which enables gentle harvesting of microtissues with control over size and composition. The method forms microtissues containing endothelial cells and mesenchymal stem cells, which are co-cultured under dynamic conditions and self-organize into blood-vessel units.

Injectable microtissue systems, devices, and methods

Fluidic connectors, methods, and devices for performing analyses (e.g., immunoassays) in microfluidic systems are provided. In some embodiments, a fluidic connector having a fluid path is used to connect two independent channels formed in a substrate so as to allow fluid communication between the two independent channels. One or both of the independent channels may be pre-filled with reagents (e.g., antibody solutions, washing buffers and amplification reagents), which can be used to perform the analysis. These reagents may be stored in the channels of the substrate for long periods amounts of time (e.g., 1 year) prior to use.

Fluidic systems for analyses

At least one microneedle comprises a hydrogel material that includes a substance that fluoresces when the substance interacts with an analyte. A magnitude of the fluorescence varies as a function of the concentration of the analyte. During use, the hydrogel material is illuminated with illumination light in a first wavelength range while the hydrogel material interfaces with the dermal interstitial fluid layer of a subject, and a photosensor generates an output that corresponds to an amount of light received in a second wavelength range.

Devices and systems for optically determining a concentration of an analyte in a living subject using hydrogel-based, fluorescent microneedles and methods of manufacture thereof

Devices systems and methods for fluid extraction and delivery to human or animal hosts are described. In embodiments, microneedles are employed to provide low infection risk and painless access to and administration of fluids. The disclosed embodiments address, among others, issues of cost, portability, ease of use in remote settings including use by untrained personnel, and others.

Fluid extraction and drug delivery system and methods using microneedles

Diagnostic systems, methods, and devices employing low-cost handheld components are disclosed herein. A diagnostic system can include a diagnostic device that is configured to perform one or more assays on a fluid sample, such as a whole blood sample, in one or more microfluidic channels or chambers. The diagnostic device can move the fluid sample into or through the one or more microfluidic channels or chambers without using any electrical power, for example, using manual actuation to generate a positive or negative pressure within the diagnostic device. The diagnostic device can have a connector for interfacing with a separate handheld unit that can provide power and data processing. For example, the separate handheld unit can be a smartphone or PDA, and the connector can interface with an existing input/output port of the unit to draw power and/or transmit data.

Diagnostic devices, systems, and methods

MicroElectroMechanical System (MEMS) devices can be fabricated completely of hydrogel materials. Such hydrogels can include polyethylene glycol with diacrylate functional groups (e.g., PEGDA), which are photopolymerizable in the presence of crosslinkers and photoinitiators. By using PEGDA monomers of different molecular weights and at different percentages, the mechanical properties of the polymerized gels and their respective permeabilities can be tuned. This spatial variation in properties and permeabilities can lead to different functionalities between different portions of the hydrogel MEMS device. Portions of the hydrogel device may be remotely actuated by applying wave energy, for example, a magnetic field, high intensity focused ultrasound, and/or infrared radiation. The remote actuation can allow the device to be actuated in vivo, for example, to allow the device to deliver a drug or other substance at a desired time and/or desired location within a patient.

Fluidic connectors and microfluidic systems

Provided are methods, pharmaceutical compositions, and kits for treatment of a metabolic condition, including obesity and type 2 diabetes by administration of a therapeutically effective amount of a cell or tissue preparation such as brown adipose microtissues to a subject, where the microtissues comprise adipose stem cells and endothelial cells. Modified approaches to creating brown adipose tissue involve differentiation of explanted white adipose tissue rather than isolation and expansion of adipose stems cells or endothelial cells and formation and differentiation of 3D cell aggregates.

An assay method is described, which comprises the steps of immobilizing a binding partner (e.g., an antigen or antibody) for an analyte to be detected (e.g., an antibody or antigen) on a portion of a surface of a microfluidic chamber; passing a fluid sample over the surface and allowing the analyte to bind to the binding partner; allowing a metal colloid, e.g., a gold-conjugated antibody, to associate with the bound analyte; flowing a metal solution, e.g., a silver solution, over the surface such as to form an opaque metallic layer; and detecting the presence of said metallic layer, e.g., by visual inspection or by measuring light transmission through the layer, conductivity or resistance of the layer, or metal concentration in the metal solution after flowing the metal solution over the surface.

Fluid delivery system and method

The present invention relates generally to microfluidic structures, and more specifically, to microfluidic structures and methods including meandering and wide channels. Microfluidic systems can provide an advantageous environment for performing various reactions and analyses due to a reduction in sample and reagent quantities that are required, a reduction in the size of the operating system, and a decrease in reaction time compared to conventional systems. Unfortunately, the small size of microfluidic channels can sometimes result in difficulty in detecting a species without magnifying optics (such as a microscope or a photomultiplier). A series of tightly packed microchannels, i.e., a meandering region, or a wide channel having a dimension on the order of millimeters, can serve as a solution to this problem by creating a wide measurement area. Although this invention mainly describes the use of meandering and wide channels in heterogeneous immunoassays on a microfluidic chip, this invention could be used for amplifying optical signals for other types of reactions and/or assays.

Fluidic structures including meandering and wide channels

A portable unitary device handheld diagnostic device can be operated with minimal power requirement and provides ease of operation as well as low cost communication of diagnostic data from remote locations. The device can provide nucleic-acid based diagnostics with minimal training, little to no sample preparation, and generates diagnostic data in about 45 minutes. A system can enable point of care transmission from any location globally using a low cost satellite-based data link technique, for example, Short Burst Data (SBD), combined with data encoding.

Field optimized assay devices, methods, and systems

Microfabricated microvalves may be used with liquid-filled control channels and actuated using compact and battery-powered components, without the need for heavier or fixed infrastructure. The disclosed embodiments include microvalves with on-off fluid control with relatively fast response times, coordinated switching of multiple valves, and operation of a biological (enzyme-substrate) assay in a handheld configuration.
