In certain embodiments, the present systems and methods use Tn7-like transposons that encode CRISPR-Cas systems for programmable, RNA-guided DNA integration. For example, the CRISPR-Cas machinery directs the Tn7 transposon-associated proteins to integrate DNA downstream of a target site (e.g., a genomic target site) recognized by a guide RNA (gRNA).

RNA-guided DNA integration using Tn7-like transposons

The present disclosure provides engineered Class 1 Type I CRISPR-Cas (Cascade) systems that comprise multi-protein effector complexes, nucleoprotein complexes comprising Type I CRISPR-Cas subunit proteins and nucleic acid guides, polynucleotides encoding Type I CRISPR-Cas subunit proteins, and guide polynucleotides. Also, disclosed are methods for making and using the engineered Class 1 Type I CRISPR-Cas systems of the present invention.

Engineered cascade components and cascade complexes

Provided are probes comprising a class 1 release factor conjugated to a fluorescent label and methods of making the probes. Also provided are methods for detecting conformational changes in ribosomes and associated molecules, such as class 1 release factors. In addition, methods of identifying a compound for reducing nonsense-mediated decay of mRNA and/or for inhibiting termination of protein synthesis at a premature stop codon, are described. Methods of assaying RF3 activity are also included.
