Provided herein are polymeric particles and compositions (i.e., 'backpacks') that can adhere to cells and provide delivery of payload immunomodulatory agents to those cells. For examples, the particles can adhere to macrophages and/or monocytes and release cytokines that promote an M1 or M2 phenotype to improve therapeutic efficacy of the cells.

Compositions and methods relating to macrophages and/or monocytes with adhered particles

Disclosed herein are methods, systems, and devices for oral drug delivery. The method generally involves orally administering a drug delivery device to the subject in need of treatment and triggering iontophoresis. The drug delivery device includes one or more active agents for delivery to the subject. The method can include the step of delivering the active agent to the intestinal mucosa after iontophoresis is triggered at the site, or simultaneously as iontophoresis is applied at the site. After the active agent(s) are delivered, the drug delivery device can be released from the intestinal mucosa. The iontophoresis can be performed for a period of time and at an electrical current that is effective to improve permeability of the one or more active agents across the intestine compared to orally administering the same drug delivery device in the absence of iontophoresis.

Oral drug delivery devices and methods using iontophoresis

Pharmaceutical compositions comprising two or more therapeutically active agents, such as two or more anticancer agents, conjugated to one or more biocompatible polymers, wherein the molar ratio of the agents and/or schedules of delivery provide a synergistic therapeutic effect, are described. Methods of making and using the pharmaceutical compositions are further described. In one embodiment, the pharmaceutical compositions contain topoisomerase I and topoisomerase II inhibitors conjugated to the same or different biocompatible polymers. The two or more anticancer agents are covalently coupled to the polymer(s), and thereby can be delivered to a tumor at a molar ratio which provides a synergistic effect. Optionally, the agents are coupled indirectly to the polymer(s) via a linker.

Polymer-drug conjugates for combination anticancer therapy

The compositions and methods described herein are topically applied to the skin with negligible or no skin irritation and can direct or prevent transport through the skin. The compositions contain neat ionic liquids, optionally in combination with a drug to be delivered. In a preferred embodiment, the compositions increase transdermal transport of the drug to be delivered. In some embodiments, the compositions disrupt bacterial biofilms. This is particularly beneficial in the treatment of antibiotic resistant skin infections. In other embodiments, the compositions direct delivery within the skin. In still other embodiments, the compositions prevent transfer of substances through the stratum corneum. The disclosed compositions and methods can be tuned and modified such that they can be used to treat or prevent a variety of different diseases and disorders.

Ionic liquids for internal delivery

Nanoscale multiple emulsions, nanoparticles resulting therefrom and methods of making and using thereof are described. The nanoscale multiple emulsions are typically double nanoemulsions, such as oil-in-water-in-oil (O1/W/O2) nanoemulsions, in which nanodroplets of the first oil (O1) phase are suspended in the aqueous (W) phase and together are dispersed in the second oil (O2) phase. The double nanoemulsions are stable with high encapsulation efficiency allowing templating to form multiphase nanoparticles. The multiphase nanoparticles include nanodroplets of O1 phase dispersed in a continuous matrix that can be a liquid, a gel, or a solid at room temperature and pressure. Methods of making the double nanoemulsions include high-energy forces and serial nanoemulsifications.

Nanoscale multiple emulsions and nanoparticles

Compounds and pharmaceutical formulations containing these compounds are described. Also described are methods of making and using the compounds. The compounds include nucleobases, nucleobase analogues, or combinations thereof. In one embodiment, a nucleobase analogue is combined with doxorubicin and encapsulated within a liposome for use in inhibiting or preventing the growth of cancer cells. Further described are pharmaceutical compositions containing two or more therapeutically active agents encapsulated within a vesicle, such as a liposome, wherein the molar ratio of the agents provides a synergistic therapeutic effect.

Drug formulations for cancer treatment

Compositions containing a complex that contains a cation with alkyl chains and a macromolecule anion, and methods of making and using are disclosed. The compositions are typically charge neutral and a liquid at room temperature and standard pressure. The macromolecule anions may be nucleic acids, peptides, proteins, and/or carbohydrates. The compositions have enhanced penetration across the skin barrier (stratum corneum) and into the skin cells, delivering the macromolecules to the skin cells. The compositions are topically applied to the skin and are particularly useful for treatment of skin conditions.

Topical formulations based on ionic species for skin treatment

Melts or ionic liquids containing amorphous propranolol, topical formulations and patches for transdermal drug delivery, and methods of making and using thereof are described herein. The melts or ionic liquids may be in a topical drug delivery formulation or patch to be applied to the skin. The drug delivery formulation or patch contains a sufficient amount of the amorphous propranolol to deliver a therapeutically effective amount of the amorphous propranolol to the patient in need of treatment, such as for the treatment or amelioration of infantile hemangioma. The formulations have a low viscosity and reduced skin irritation compared to the crystalline propranolol free base (PFB). The melts or ionic liquids can be formed by a salt metathesis reaction.

Formulations of propranolol and analogs as an amorphous melt or ionic liquid for transdermal drug delivery

Provided herein are novel hyaluronic acid-based materials for in-situ gelation in wounds and other contexts, which materials are biocompatible and which have favorable gelation kinetics. The resulting hydrogels have desirable rheological properties. The invention encompasses novel reagents for forming crosslinked hyaluronic acid hydrogels, with favorable kinetics for medical applications and having desirable and tunable physical properties. Also presented herein are methods of using the novel materials disclosed herein in wound care and other medical contexts.

Hyaluronic acid-based hydrogels having medical applications

The current invention provides methods, as well as compositions useful in such methods, involving application of energy to a tissue of interest to generate a liquefied sample comprising tissue constituents so as to provide for rapid tissue sampling, as well as qualitative and/or quantitative detection of analytes that may be part of tissue constituents (e.g., several types of biomolecules, drugs, and microbes). Determination of tissue composition can be used in a variety of applications, including diagnosis or prognosis of local as well as systemic diseases, evaluating bioavailability of therapeutics in different tissues following drug administration, forensic detection of drugs-of-abuse, evaluating changes in the tissue microenvironment following exposure to a harmful agent, and various other applications.
