This flow cytometer includes a flow path through which an observation object flows with a fluid; an optical illumination system including a spatial optical modulation device, and a first optical element; and an optical detection system including a first light detector, wherein the optical illumination system further includes a first spatial filter disposed in a first optical path between a light source and an image position of light imaged in the flow path by the first optical element and having a first region which hinders traveling of light emitted from the light source towards the observation object, the optical detection system further includes a second light detector disposed in a second optical path between the first light detector and the image position and having a second region which directs the light modulated by the observation object towards the first light detector, and the position of the first region and the position of the second region are in a substantially optically conjugate relationship.

Flow cytometer

An observation device includes an illumination optical system and an observation optical system. The illumination optical system includes a light source and an aperture member. The observation optical system includes an objective lens, an optical structure, and a detector. The optical structure is disposed at a first position which is the position conjugate with the aperture member. The optical structure includes a blocking portion that blocks light and a transmitting portion that transmits light, the blocking portion having a shape including the shape of an image of an aperture of the aperture member which is formed on the optical structure. The detector detects dark-field light passing through the optical structure.

Observation device

An imaging flow cytometer includes: a flow channel in which an observation object flows and a length in a width direction is longer than a length in a height direction; an acoustic element configured to apply acoustic waves as standing waves to the flow channel; a light source that irradiates the flow channel with illumination light; an image sensor configured to image at least a line included in a cross section of the observation object crossing a flow line direction which is a direction in which the observation object flows in the flow channel by measuring or imaging the observation object passing through a position irradiated with the illumination light; and circuitry configured to generate an image in which the observation object is scanned in the flow line direction on the basis of a plurality of captured images acquired by the imaging unit imaging the line in a time series.

Imaging flow cytometer

The present disclosure provides methods and systems for ghost cytometry (GC), which may be used to produce an image of an object without using a spatially resolving detector. This may be used to perform image-free ultrafast fluorescence 'imaging' cytometry, based on, for example, a single pixel detector. Spatial information obtained from the motion of cells relative to a patterned optical structure may be compressively converted into signals that arrive sequentially at a single pixel detector. Combinatorial use of the temporal waveform with the intensity distribution of the random or pseudo-random pattern may permit computational reconstruction of cell morphology. Machine learning methods may be applied directly to the compressed waveforms without image reconstruction to enable efficient image-free morphology-based cytometry. Image-free GC may achieve accurate and high throughput cell classification as well as selective sorting based on cell morphology without a specific biomarker, which have been challenging using conventional flow cytometers.

Methods and systems for cytometry

An analysis device includes an analysis unit configured to receive scattered light, transmitted light, fluorescence, or electromagnetic waves from an observed object located in a light irradiation region light-irradiated from a light source and analyze the observed object on the basis of a signal extracted on the basis of a time axis of an electrical signal output from a light-receiving unit configured to convert the received light or electromagnetic waves into the electrical signal.

Analysis device

An electromagnetic wave detecting device comprising: an emission unit configured to emit electromagnetic waves having coherence; an electromagnetic wave modulating unit configured to modulate one or both of a phase and an amplitude of the emitted electromagnetic waves and to change a state of the modulation relative to an imaging target; and a post-modulation electromagnetic wave intensity detecting unit configured to detect an intensity of post-modulation electromagnetic waves, which are the modulated electromagnetic waves acquired by modulating the electromagnetic waves emitted from the emission unit using the imaging target and the electromagnetic wave modulating unit, using one pixel.

Any one or both of an optical system with a structured lighting pattern and a structured detecting system having a plurality of regions with different optical characteristics are used. In addition, optical signals from an object to be observed through one or a small number of pixel detectors are detected while changing relative positions between the object to be observed and any one of the optical system and the detecting system, time series signal information of the optical signals are obtained, and an image associated with an object to be observed from the time series signal information is reconstructed.

The present disclosure provides a method for screening cells, the method including a step of preparing a plurality of cells which are tagged with a first barcode nucleic acid associated with a test target and treated with the test target, a step of sorting the plurality of cells based on cellular phenotype using an imaging cell sorter, and a step of identifying the test target used to treat each cell using the first barcode nucleic acid as an indicator.

An imaging flow cytometer includes at least one flow channel through which an observation target flows, a light source which irradiates the flow channel with sheet-like excitation light, an imaging unit which images a specific cross-section of the observation target by imaging fluorescence from the observation target having passed through a position irradiated with the excitation light, and a three-dimensional image generation unit which generates a three-dimensional image of the observation target as a captured image on the basis of a plurality of captured images obtained by cross-sectional imaging by the imaging unit.
