There is disclosed a lactam derivative of the formula: ##STR1## wherein R.sup.1 is a substituted or unsubstituted phenyl group; R.sup.2 is a substituted or unsubstituted phenyl group, a cycloalkyl group or a nitrogen-containing 6-membered heterocyclic group; Z is oxygen atom or sulfur atom; and n is an integer of 2 or 3, and a salt thereof. The above lactam derivative is novel and useful as a pharmaceutical compound.

An anti-vertigo drug which comprises as an active ingredient 6-amino-2-fluoromethyl-3-(o-tolyl)-4(3H)-quinazolinone (Afloqualone) or a pharmaceutically acceptable acid addition salt thereof, which is effective for the prophylaxis and treatment of vertigo and also such various symptoms accompanied with an abnormality in the reflex system of vestibulo-equilibrium sense as desesthesia of motion and position, nystagmus, dysequilibrium, head deviation, nausea, vomiting, sweating, salivation and tachycardia.

N-(2-amino-5-nitrobenzoyl)-o-toluidine is condensed with fluoroacetic acid or a functional derivative thereof. N-(2-fluoroacetamido-5-nitrobenzoyl)-o-toluidine obtained by said condensation reaction is dehydrated to produce 2-fluoromethyl-3-(o-tolyl)-6-nitro-4(3H)-quniazolinone, which is then subjected to reduction reaction. 2-fluoromethyl-3-(o-tolyl)-6-amino-4(3H)-quinazolinone thus obtained and a pharmaceutically acceptable acid addition salt thereof are useful as central muscle relaxants, minor tranquilizers, anti-convulsants, analgesics and/or anti-inflammatory agents.
