Beads with functionalized material applied to them are exposed to an acoustic field to trap, retain or pass the beads. The beads may include or be free of ferro magnetic material. The beads may be biocompatible or biodegradable for a host. The size of the beads may vary over a range, and/or be heterogenous or homogenous. The composition of the beads may include high, neutral or low acoustic contrast material. The chemistry of the functionalized material may be compatible with existing processes. The acoustic field may be generated, for example, in an acoustic angled wave device or in an acoustic fluidized bed.

Acoustic cell separation techniques and processes

Cell therapy processes utilizing acoustophoresis

Acoustophoretic devices are disclosed. The devices include a flow chamber, an ultrasonic transducer, a reflector, an inlet, a filtrate outlet, a concentrate outlet, and optionally a lipid collection trap. The ultrasonic transducer and reflector create a multi-dimensional acoustic standing wave in the flow chamber that traps and separates red blood cells and/or lipids from blood. Concentrated red blood cells can be recovered via the concentrate outlet, the lipids can be recovered via the lipid collection trap, and the remaining blood can be recovered via the filtrate outlet. Methods for separating blood components (e.g., red blood cells, lipids, platelets, white blood cells) from blood are also disclosed. The red blood cells can undergo washing with a solvent to remove undesired admixtures. Cryoprotectants can be added or removed from the blood.

Acoustic blood separation processes and devices

An acoustophoretic device is disclosed. The acoustophoretic device includes an acoustic chamber, an ultrasonic transducer, and a reflector. The ultrasonic transducer includes a piezoelectric material driven by a voltage signal to create a multi-dimensional acoustic standing wave in the acoustic chamber emanating from a non-planar face of the piezoelectric material. A method for separating a second fluid or a particulate from a host fluid is also disclosed. The method includes flowing the mixture through an acoustophoretic device. A voltage signal is sent to drive the ultrasonic transducer to create the multi-dimensional acoustic standing wave in the acoustic chamber such that the second fluid or particulate is continuously trapped in the standing wave, and then agglomerates, aggregates, clumps, or coalesces together, and subsequently rises or settles out of the host fluid due to buoyancy or gravity forces, and exits the acoustic chamber.

Non-planar and non-symmetrical piezoelectric crystals and reflectors

Methods are disclosed for separating beads and cells from a host fluid. The method includes flowing a mixture containing the host fluid, the beads, and the cells through an acoustophoretic device having an ultrasonic transducer including a piezoelectric material driven by a drive signal to create a multi-dimensional acoustic standing wave. A drive signal is sent to drive the at least one ultrasonic transducer to create the multi-dimensional acoustic standing wave. A recirculating fluid stream having a tangential flow path is located substantially tangential to the standing wave and separated therefrom by an interface region. A portion of the cells pass through the standing wave, and the beads are held back from the standing wave in the recirculating fluid stream at the interface region. Also disclosed is an acoustophoretic device having a coolant inlet adapted to permit the ingress of a cooling fluid into the device for cooling the transducer.

Acoustic perfusion devices

Multi-stage acoustophoretic devices for continuously separating a second fluid or a particulate from a host fluid are disclosed. Methods of operating the multi-stage acoustophoretic devices are also disclosed. The systems may include multiple acoustophoretic devices fluidly connected to one another in series, each acoustophoretic device comprising a flow chamber, an ultrasonic transducer capable of creating a multi-dimensional acoustic standing wave, and a reflector. The systems can further include pumps and flowmeters.

Therapeutic cell washing, concentration, and separation utilizing acoustophoresis

Acoustophoretic devices and methods for separating biological cells (particularly T-cells) from other fluids/materials using multi-dimensional acoustic standing waves are disclosed. The devices include an inlet, at least two outlets, and a flow chamber having an ultrasonic transducer-reflector pair. Specifically, T cells, B cells, or NK cells can be separated from other blood components. A dual-pass acoustophoretic system including two acoustophoretic devices arranged in series and fluidly connected to one another is also illustrated. Means for pre-chilling the mixture prior to separation in the devices or system can be used to improve retention, concentration, and clarification and to prevent outgassing.

Acoustic separation of T cells

Devices and methods for inspecting, detecting, isolating, monitoring, characterizing, or separating pathogens in blood containing blood cells are disclosed. The devices include a flow chamber having a solvent inlet, at least one host-fluid inlet, a particulate outlet, at least one residual outlet, and a reflector. The methods include trapping the pathogens in the acoustic standing wave, introducing a solvent into the flow chamber, and removing the pathogens from the device. Devices and methods for inspecting, detecting, isolating, monitoring, characterizing, or separating specialized circulating cells in blood containing blood cells are also disclosed. The devices include a flow chamber having at least one inlet and at least one outlet, and a microscope objective and a cover glass. The methods include driving the transducer to create an acoustic standing wave in the flow chamber and microbubbles in the blood.

Acoustic methods for separation of cells and pathogens

Methods, systems, and apparatus for acoustic separation of cellular supporting materials such as microcarriers or microbubbles from cell culture are provided. In one aspect, a method includes flowing a fluid containing the cellular supporting material and the cells through a flow chamber; driving at least one acoustic transducer to launch an acoustic wave from the acoustic transducer positioned on a first wall of the flow chamber to a reflector positioned on a second wall of the flow chamber to create, in the flow chamber, a multi-dimensional field that includes first spatial locales where acoustic pressure amplitude is elevated from a baseline level when the acoustic transducer is turned off, and second spatial locales where acoustic pressure amplitude is substantially identical to the baseline level when the acoustic transducer is turned off, wherein the first wall is opposite to the second wall; preferentially trapping, and gravity separating, the cellular supporting material or the cells at the first or second spatial locales inside the flow chamber; and collecting cells separated from the cellular supporting material using a first collection duct coupled to the flow chamber.

Acoustic separation of cellular supporting materials from cultured cells

A chamber in which an agent, like genomic DNA, may be harvested and optionally manipulated rapidly (e.g., on the order of a few hours), without shearing or fragmentation.

Reaction chamber

The invention provides methods and systems for improved spatial resolution of signal detection, particularly as applied to the analysis of polymers such as biological polymers. Some of the methods and systems comprise differentially tagging polymers in order to increase resolution. Some of the methods and systems comprise techniques for improving the precision of separation distance measurements, without necessarily requiring improvements in the known detection resolution of prior art systems.
