Superparamagnetic nanoparticle-based analytical method comprising providing a sample having analytes in a sample matrix, providing a point of care chip having analytical regions, each of which is a stationary phase having at least one or more sections, labeling each of the analytes with a superparamagnetic nanoparticle and immobilizing the labeled analytes in the stationary phase, providing an analytical device having a means for exciting the superparamagnetic nanoparticles in vitro and a means for sensing, receiving, and transmitting response of the excited superparamagnetic nanoparticles, placing the chip in the analytical device and exciting the superparamagnetic nanoparticles in vitro, sensing, receiving, and transmitting the response of the superparamagnetic nanoparticles, and analyzing the response and determining characteristic of the analytes, wherein the response of the superparamagnetic nanoparticles comprises harmonics. The present invention also provides the hybrid point of care chip and analyzer to be used in the analytical method.

Superparamagnetic particle imaging and its applications in quantitative multiplex stationary phase diagnostic assays

An improved apparatus and method for dispersion of a labeling conjugate in a diagnostic assay, the result being a one-step assay. By eliminating a conjugate pad as in conventional lateral diagnostic devices, and forming a frazil ice pellicle (FIP), rehydration and flow are improved resulting in better reproducibility, improved sensitivity, and reduced costs of individual assay devices. The formation of a frazil ice film formed on a super cooled surface of a sample receiving means simplifies assay assembly. Lyophilization of the FIP improves the release of a sample/analyte/label matrix into a macro channel as in a direct flow assay, while at the same time allowing reagents to mix and flow, thereby optimizing the assay performance. The reagents of the conjugate and the formation of the FIP stabilize the conjugate proteins and provide extended shelf life to the diagnostic assay device.

Frazil ice conjugate assay device and method

A diagnostic assay device that directs an applied sample to the analytical membrane of a directed flow device. The device has a sample receiving port defined by layers of built-up material on one end of the test strip. The port contains the sample and specifically directs it to the membrane in a controlled fashion. Additional features include configuration of the housing in a general C-shape with the test strip spanning the opening of the C-shape to allow access by a reader device. A preferred method employs superparamagnetic particles to label the target analytes for detection and measurement by means of an electromagnetic reader device.

Directed-flow assay device

An assay test strip and cassette. The test strip is positioned in a housing shaped and configured to allow a detector to access the test strip from the side, rather than from the lengthwise axis of the test strip. The housing may contain one or more test strips which may also be disposed on one or more surfaces of the housing. Preferably, the housing is generally C-shaped with the test strip spanning the space between the two arms of the C-shape. The housing is sealed to protect both the operator and instrument from possible contamination. The test strip is preferably embedded with paramagnetic particles and process chemistry specific for a particular application. Quantitative analysis may be accomplished using a magnetic reader device. In additional embodiments, detection is accomplished by visual means.

Accessible assay and method of use

An immunochromatographic assay employing superparamagnetic particles to lable the target analytes. An opaque cover prevents misinterpretive readings in field situations and provides a protective surface on the porous membrane. Additional features include separability of the test strip from any backing or housing which is configured to support the strip, and that quantitative measurements of the target analytes are easily and accurately made by means of an electromagnetic reader device.
