There are provided herein, inter alia, complexes, compositions and methods for the delivery of nucleic acid into a cell in vivo. The complexes, compositions and methods may facilitate complexation, protection, delivery and release of oligonucleotides and polyanionic cargos into target cells, tissues, and organs both in vitro and in vivo.

Hybrid immolative cell-penetrating complexes for nucleic acid delivery

There are provided herein, inter alia, cationic amphipathic polymers, complexes, and compositions comprising same, and methods for their use including for the delivery of therapeutic, diagnostic and imaging agents, including nucleic acids, into a cell. The complexes, compositions and methods may facilitate delivery and targeted release of the therapeutic, diagnostic and imaging agents to particular cell types and tissues.

Immolative cell-penetrating complexes for nucleic acid delivery to the lung

Humanized or chimeric anti-CD81 (cluster of differentiation 81) monoclonal antibodies are provided. The antibodies bind to human CD81, and find use in various therapeutic methods, including without limitation the reduction or prevention of tumor metastasis. Further provided are heavy chain and light chain variable region sequences as well as associated complementarity-determining region (CDR) sequences.

Methods for enhancing anti-tumor antibody therapy

Humanized or chimeric anti-CD99 monoclonal antibodies are provided. The antibodies bind to and neutralize human CD99, and find use in various therapeutic methods. Embodiments of the invention include isolated antibodies and derivatives and fragments thereof, pharmaceutical formulations comprising one or more of the humanized or chimeric anti-CD99 monoclonal antibodies; and cell lines that produce these monoclonal antibodies. Also provided are amino acid sequences of the antibodies.

Predictive biomarkers identify those patients suffering from immunoglobulin positive (Ig) B lineage malignancies that are responsive to active immunotherapy, where the active immunotherapy comprises vaccination with a tumor-specific idiotype-immunogen. It is shown herein that patient responsiveness to the idiotype-immunogen is dependent upon the sequence of the immunogen, where an immunogen having a low number of tyrosine residues in the CDR1 (herein termed CDR1-Y) regions of one or both of the immunogen heavy and light chains is predictive of a positive anti-tumor response, while a high number of CDR1 tyrosine residues (herein termed CDR1-Y) is predictive of a low anti tumor response.

Method of predicting responsiveness of B cell lineage malignancies to active immunotherapy

Methods and compositions are provided for determining whether a subject suffering from a neoplastic condition is responsive to an antineoplastic therapy, such as antibody therapy, e.g., Rituximab. In practicing the subject methods, the subject is genotyped to determine whether the subject has a least one favorable Fc&#x3b3;R polymorphism, e.g., the 131 H/H genotype or the 158 V/V genotype. In addition, reagents, devices and kits thereof, that find use in practicing the subject methods are provided.

Methods and compositions for determining responsiveness to antibody therapy

Methods of enhancing the efficacy of antibody-directed cellular cytotoxicity (ADCC) for therapy directed to killing of tumor cells are disclosed. Cancer specific cell surface antigens are bound by monoclonal antibodies, thereby stimulating a cytotoxic T cell response characterized by an upregulation of cell surface expression of costimulatory molecules on the T cell. The ADCC response is augmented by the subsequent administration of a second antibody that is an agonist of the costimulatory molecule.

Methods and kits for classifying patients having diffuse large B-cell lymphoma (DLBCL) based upon expression of a plurality of genes are disclosed. Real-time quantitative RT-PCR can be used to measure expression values. Methods and composition are provided for identifying antiproliferative polypeptides which inhibit clonal expansion and/or induce apoptosis in cells of a predetermined cell population (e.g., a neoplastic cell sample) expressing a cell surface receptor which is a member of the immunoglobulin superfamily. A predetermined cell population expressing surface immunoglobulin superfamily molecules is isolated from a patient as a cellular sample, such as a lymph node biospy or blood sample containing neoplastic lymphocytic cells. Antiproliferative peptides which are identified by the methods of the invention can be used as therapeutic agents for treating lymphoproliferative disorders by anti-idiotype therapy.
