A bioartificial nucleus pulposus is adapted for replacing at least a portion of a natural nucleus pulposus in an intervertebral disc. The bioartificial nucleus pulposus includes an artificial support structure made from a biodegradable material. The support structure has a plurality of pores. A carrier material is contained in the pores of the support structure. A plurality of nucleus pulposus cells are carried by the carrier material. A bioartificial intervertebral disc contains the bioartificial nucleus pulposus.

Bioartificial intervertebral disc

A system and method is provided for optimizing a laser light fractionation during photodynamic therapy of a tumor. The tumor is destroyed through the production of singlet oxygen and successful photodynamic therapy treatment requires the maintenance of tumor oxygen above a specified critical level. Depending on the tumor, the photosensitizer concentration, and the laser fluence, sustained irradiation of the tumor will deplete the oxygen below the critical level. When the laser is turned off, oxygen diffuses into the tumor to provide sustained levels of singlet oxygen. The control system controls the operation cycle of the laser and the various surgical parameters. Data regarding the sensitizer, the laser characteristics, and the oxygen features of the tumor are input into the control system for simulation of the PDT treatment. As part of the simulation, the optimal fractionation cycle for the laser is predicted. The data may be adjusted for conducting additional simulations. The simulation data and results are stored in the control system for controlling the actual PDT treatment cycle. A computer is linked to the laser and a beam splitter or other cycling device to control the fractionated photodynamic therapy irradiation process for optimal treatment of a specific tumor.

An implantable bioartificial pancreas device having an islet chamber containing glucose responsive and insulin-secreting islets of Langerhans or similar hormone secreting cells, one or more vascularizing chambers open to surrounding tissue, a semi-permeable membrane between the islet and vascularizing chambers that allows passage of small molecules including insulin, oxygen and glucose and does not allow passage of agents of the immune system such as white cells and antibodies, the vascularizing chambers containing a growth factor soaked fibrous or foam matrix having a porosity of about 40 to 95%, the matrix providing small capillary growth and preventing the blood from clotting in the lower chamber.

Bioartificial pancreas

A method of providing and sustaining a difference in pH of a first reaction of an immobilized enzyme and a second reaction in a bulk liquid surrounding the immobilized enzyme is carried out with a bilayer pellet containing coimmobilized enzymes. The pellet can contain an enzyme that produces a desired product immobilized in an inner core and urease immobilized in an outer layer. The bulk liquid contains urea and a substrate for the enzyme in the core, and has an acidic pH. The urease reacts with urea diffusing into the outer layer from the bulk liquid to produce ammonia. The ammonia consumes hydrogen ions diffusing into the inner core from the acidic bulk liquid. This provides the enzyme in the inner core with a pH higher than the acidic pH of the bulk liquid suitable for the enzyme to react with the substrate as it diffuses into the inner core. In a preferred embodiment, simultaneous isomerization of xylose to xylulose and fermentation of xylulose to ethanol is carried out with a bilayer pellet containing xylose isomerase in the core and yeast in the bulk liquid. The isomerization occurs at an optimum pH of 7.0 to 8.0 and the fermentation occurs at an optimum pH of 4.0 to 5.0.

A bilayered immobilized enzyme pellet and a process to manufacture this pellet are provided for use in a process involving the simultaneous isomerization of xylose to xylulose and fermentation of xylulose to ethanol. The bilayered pellet is able to maintain the environment where the isomerization reaction occurs within its optimum pH of 7.0 to 8.0 while the fermentation reaction occurs within its optimum pH range of 4.0 to 5.0. This process allows both xylose and glucose sugars to be effectively used as a feedstock for ethanol production by isomerizing the xylose to xylulose and then making the xylulose immediately available for the fermentation process. Because the xylose has been converted to its ketose isomer, xylulose, yeasts which can ferment glucose and xylulose can be used in this process.
