Systems and methods of using the same for functional fluorescence imaging of live cells in suspension with isotropic three dimensional (3D) diffraction-limited spatial resolution are disclosed. The method-live cell computed tomography (LCCT)-involves the acquisition of a series of two dimensional (2D) pseudo-projection images from different perspectives of the cell that rotates around an axis that is perpendicular to the optical axis of the imaging system. The volumetric image of the cell is then tomographically reconstructed.

Live-cell computed tomography

Systems and methods of using the same for functional fluorescence imaging of live cells in suspension with isotropic three dimensional (3D) diffraction-limited spatial resolution are disclosed. The method-live cell computed tomography (LCCT)-in-volves the acquisition of a series of two dimensional (2D) pseudo-projection images from different perspectives of the cell that rotates around an axis that is perpendicular to the optical axis of the imaging system. The volumetric image of the cell is then tomographically reconstructed.

A microfluidic device useable for performing live cell computed tomography imaging is fabricated with a cover portion including a first wafer with at least one metal patterned thereon, a base portion including a second wafer with at least one metal patterned thereon and negative photoresist defining recesses therein, and a diffusive bonding layer including a negative photoresist arranged to join the cover portion and the base portion. A composition useful in live cell computer topography includes a long-chain polysaccharide at a concentration of from about 0.01% to about 10.0% in cell culture medium for supporting cell life while enabling cell rotation rate to be slowed to a speed commensurate with low light level imaging.

Microfluidic electrocage device and cell medium for trapping and rotating cells for live-cell computed tomography (CT)

Microfluidic devices for 3D hydrodynamic microvortical rotation of at least one live single cell or cell cluster, systems incorporating the devices, and methods of fabricating and using the devices and systems, are provided. A microfluidic chip rotates at least one live single cell or cell cluster in a microvortex about a stable rotation axis perpendicular to an optical axis within a chamber having a trapezoidal cross-sectional shape located below a flow channel. An optical trap may be used to position the cell or cells with the microvortex, and the cell or cells may be subject to live-cell or cell cluster computer tomography imaging.

Microfluidic systems and methods for hydrodynamic microvortical cell rotation in live-cell computed tomography

Novel methods and devices including modified endoscopes that employ fiber optic technology and multiple extrinsic optical sensors for enablement of simultaneous determination of change rates of biological analytes in vivo with single-cell resolution. The devices employ dynamic oscillatory actuation to determine the gradient of analytes of interest along one or multiple directions with respect to the cell. The change rate or flux of the analyte of interest can be easily determined by applying the first Fick's law that relates the flux with the concentration gradient. The dual operation mode of the device markedly increases measurement sensitivity and accuracy.

Endoscope for analyte consumption rate determination with single cell resolution for in vivo applications

This disclosure relates generally to potassium ion sensors and monomers derived from such sensors. The disclosure also provides for polymers (e.g., random copolymers, nanoparticles, polymer thin films, and sensors) having polymerized monomeric potassium ion sensors as described herein. These compounds and polymers are useful for measuring intracellular and extracellular potassium ion concentrations.

Fluorescent potassium ion sensors

An optical tomography system for imaging an object of interest including a light source for illuminating the object of interest with a plurality of radiation beams. The object of interest is held within an object containing tube such that it is illuminated by the plurality of radiation beams to produce emerging radiation from the object containing tube, a detector array is located to receive the emerging radiation and produce imaging data used by a mechanism for tracking the object of interest.

Focal plane tracking for optical microtomography

A parallel-beam optical tomography system for imaging an object of interest includes a parallel ray beam radiation source that illuminates the object of interest with a plurality of parallel radiation beams. After passing through the object of interest the pattern of transmitted or emitted radiation intensities is magnified by a post specimen optical element or elements. An object containing tube is located within an outer tube, wherein the object of interest is held within or flows through the object containing tube. A motor may be coupled to rotate and/or translate the object containing tube to present differing views of the object of interest. One or more detector arrays are located to receive the emerging radiation from the post specimen magnifying optic. Two- or three-dimensional images may be reconstructed from the magnified parallel projection data.

Optical tomography of small objects using parallel ray illumination and post-specimen optical magnification

A microtomographic system (10) for generating high-resolution, three dimensional images of a specimen (16) is disclosed. The microtomograph system includes an x-ray generator (12) that produces an x-ray beam (14), a specimen holder (18) that holds the specimen in the beam, and an x-ray detector (20) that measures the attenuation of the beam through the specimen. Two projections of each view of the specimen are made with this microtomographic system. Each projection is made with a different intensity x-ray beam. After the projections of one view of the specimen are made, the specimen is rotated on the specimen holder and another set of projections are made. The projections of each view of the specimen are analyzed together to provide a quantitative indication of the phase fraction of the material comprising the specimen. The projections of the different views are combined to provide a three-dimensional image of the specimen.

Three-dimensional microtomographic analysis system

A femoral-side hip-joint prosthetic device for use in hip-joint replacement. The device includes an elongate stem whose surface defines maximally spaced infero-medial and supero-lateral surface curves, and which is characterized by a surface rotation which carries the supero-lateral surface curve about 15.degree.-30.degree. forward with respect to the infero-medial surface curve, on progressing upward along the upper surface portion of the stem, with the device in operative position. The rotation of the stem surface produces a close approximation to the cavity formed in the proximal femur by removal of the cancellous bone in the natural cavity formed by a wall of densified cancellous bone. A 3.degree.-15.degree. proximal anteversion of the neck is adapted to place a hip-joint ball carried on the stem at a position closely approximating that of the head in the natural femur. In the method of the invention, the cavity which receives the prosthesis stem is formed by a rasp/broach-type tool whose surface conforms closely to that of the stem.

Femoral-joint prosthetic device

An improved prosthesis construction in a femoral-side hip joint replacement device of the type having an elongate curved stem held within an intramedullary cavity in a femur, and a neck supporting a ball-like joint member at the femur's proximal end. The construction includes an elongate polymer core containing continuous-filament fibers oriented substantially along the length of the core. The core contains the neck, an elongate distal stem whose cross-sectional area is less than about one-quarter that of the neck, and a tapered sections which mates the neck to the stem. A polymer skin fused to and covering the stem and tapered section of the core is shaped and dimensioned to conform to and fill the bone cavity. A braided sheath encases the stem and tapered sections of the core and it embedded in the polymer skin adjacent the core. The filaments which made up the braid in the sheath encircle the core in a helical pattern extending along the stem and tapered section provided resistance to torsional stresses on the stem. Also disclosed are novel methods for making the prosthesis construction.
