The present invention generally relates to combination immunoassays, reagents and kits for simultaneous detection of HCV antigens and anti-HCV antibodies in a test sample.

HCV antigen-antibody combination assay and methods and compositions for use therein

The present disclosure relates to polypeptides, including fusions thereof, nucleic acids, vectors, host cells, immunodiagnostic reagents, kits, and immunoassays for use detecting the presence of HCV antibodies. More specifically, the present invention describes specific NS3 antigens that can be used for the detection of anti-HCV antibodies.

HCV NS3 recombinant antigens and mutants thereof for improved antibody detection

A polypeptide comprising the contiguous amino acids 1-198 of SEQ ID NO: 2; a polypeptide, which comprises a contiguous amino acid sequence that is at least about 95% identical to the contiguous amino acids 1-198 of SEQ ID NO: 2, an epitope that is immunoreactive with an antibody that specifically binds to the core protein of hepatitis C virus (HCV), and an epitope that is immunoreactive with an antibody that specifically binds to the NS4 region of HCV; a nucleic acid encoding such a polypeptide; a host cell comprising such a nucleic acid; an immunodiagnostic reagent comprising such a polypeptide; a kit comprising such an immunodiagnostic reagent; and a method of determining the presence, amount, or concentration of anti-HCV antibodies in a test sample.

Materials and methods for assay of anti-hepatitis C virus (HCV) antibodies

Recombinant antibodies, including chimeric antibodies, specific for hepatitis C (HCV) antigenic proteins are provided. The recombinant antibodies specifically bind to diagnostically relevant regions of HCV proteins and are thus suitable for use, for example, as diagnostic reagents for the detection of HCV, and/or as standardization reagents or positive control reagents in assays for the detection of HCV. The recombinant antibodies can also be used in the treatment or prevention of a HCV infection.

Recombinant antibodies against hepatitis C virus and methods of obtaining and using same

The subject invention relates to methods for the simultaneous detection of Hepatitis C Virus (HCV) antigens as well as antibodies produced in response to HCV antigens. Furthermore, the subject invention allows one to detect antigens in the early, acute stage of infection, even prior to the development of antibodies, thereby allowing for early detection of infected blood and blood products, thus improving the safety of the blood supply.

The present invention also provides an assay for detecting the presence of an antibody to an HCV antigen in a sample by contacting the sample with the recombinant antigens. Preferred assay formats include a screening assay, a confirmatory assay, a competition or neutralization assay and an immunodot assay.

The present invention provides unique recombinant antigens representing distinct antigenic regions of the Hepatitis C Virus (HCV) genome which can be used as reagents for the detection of antibodies and antigen in body fluids from individuals exposed to HCV.
