A method and device for preferentially delivering a compound such as an antigen to the cytosol of an immune cell. The method comprises passing a cell suspension comprising the target immune cell through a microfluidic device and contacting the suspension with the compound(s) or payload to be delivered.

Delivery of biomolecules to immune cells

The present invention provides inventive conjugated polyethyleneimine (PEI) polymers and conjugated aza-macrocycles (collectively referred to herein as “conjugated lipomers” or “lipomers”) containing one or more groups of the formula (iii): wherein Rand Rare as defined herein. Conjugated lipomers and uses thereof

Drug delivery articles, resident articles, and retrieval systems e.g., for gram-level dosing, are generally provided. In some embodiments, the articles are configured for transesophageal administration, transesophageal retrieval, and/or gastric retention to/in a subject. In certain embodiments, the article includes dimensions configured for transesophageal administration with a gastric resident system. In some cases, the article may be configured to control drug release e.g., with zero-order drug kinetics with no potential for burst release for weeks to months. In some embodiments, the articles described herein comprise biocompatible materials and/or are safe for gastric retention. In certain embodiments, the article includes dimensions configured for transesophageal retrieval. In some cases, the articles described herein may comprise relatively large doses of drug (e.g., greater than or equal to 1 gram).

Resident articles for gram-level dosing

Self-actuating articles including, for example, self-actuating needles and/or self-actuating biopsy punches, are generally provided. Advantageously, the self-actuating articles described herein may be useful as a general platform for delivery of a wide variety of pharmaceutical drugs that are typically delivered via injection directly into tissue due to degradation in the GI tract. The self-actuating articles described herein may also be used to deliver sensors and/or take biopsies without the need for an endoscopy. In some embodiments, the article comprises a spring (e.g., a coil spring, a beam, a material having particular mechanical recovery characteristics). Those of ordinary skill in the art would understand that the term spring is not intended to be limited to coil springs, but generally encompass any reversibly compressive material and/or component which, after releasing an applied compressive force on the material/component, the material/component substantially returns to an uncompressed length of the material/component (e.g., the within 95% of the length of the material/component prior to compression).

Self-actuating articles

Described herein are methods such as multi-omic methods for assessing a disease such as cancer. The multi-omic methods may integrate proteomic, transcriptomic, genomic, lipidomic, or metabolomic data. The method screening diseases or disease states. Also described herein are methods for screening for diseases or disease states from biological samples. The methods may include assessing whether a nodule, mass, or cyst is cancerous.

Multi-omic assessment using proteins and nucleic acids

Compositions, articles, and methods for targeted drug delivery, such as thermoresponsive hydrogel polymers, are generally provided. In one aspect, the compositions and articles comprise a thermoresponsive hydrogel polymer comprising a releasable therapeutic agent. In some cases, the compositions described herein have advantageous combinations of properties including mechanical strength, biocompatibility, tunable charge densities, thermal responsiveness, drug loading, and/or configurations for targeted drug delivery. In one embodiment, the composition comprises a solution comprising a thermoresponsive polymer including one or more ligands attached to the polymer, wherein the solution is configured to undergo a sol-to-gel transition under physiological conditions. In another embodiment, the composition comprises a plurality of nanoparticles e.g., associated with the thermoresponsive polymer. In yet another embodiment, the composition comprises a therapeutic agent e.g., associated with the nanoparticles and/or thermoresponsive polymer.

Compositions and related methods for targeted drug delivery

Gastric resident electronics, devices, systems, and related methods are generally provided. Some embodiments comprise administering (e.g., orally) an (electronic) resident structure to a subject (e.g., a patient) such that the (electronic) resident structure is retained at a location internal to the subject for a particular amount of time (e.g., at least about 24 hours) before exiting said location internal to the subject. In some embodiments, the resident structure is a gastric resident electronic. That is to say, in some embodiments, the resident structure is configured for relatively long gastric residence and comprises an electronic component. In some embodiments, the structures and components described herein may comprise one or more components configured for the delivery of an active substance(s) (e.g., a pharmaceutical agent) to the subject. In some embodiments, the device has a modular design, combining an electronic component(s) with materials configured for controlled and/or tunable degradation/dissolution to determine the time at which (gastric) residence is lost and the device exits the location internal to the subject. For example, in some embodiments, the resident structure comprises an electronic component and one or more additional components associated with the electronic component such that the resident structure is configured to be retained at a location internal to a subject for greater than or equal to 24 hours.
