Compositions and methods are presented that provide substantially improved physical and pharmacological parameters for subungually administrable formulations. Most beneficially, the compositions presented herein have a viscosity and film-forming capacity that retain the liquid formulation in sufficient quantities and that help penetrate the polysaccharide matrix commonly associated with onychomycosis. In especially preferred aspects, the carrier is formulated to have a high concentration of the active pharmaceutically active agent (API), to allow migration of the formulation within the subungual space, and to reduce systemic absorption while promoting diffusion of the API into the nail plate at or above minimum inhibitory concentration into a larger treatment space.

Onychomycosis treatment compositions and methods

Disclosed herein are pharmaceutical compositions and dosage forms including N-[5-(3,5-difluorobenzyl)-1H-indazol-3-yl]-4-(4-methyl-piperazin-1-yl)-2-(tetrahydro-2H-pyran-4-ylamino) benzamide that are useful in the treatment of subjects having cancer. The present disclosure also provides methods for preparing these pharmaceutical compositions and dosage forms, and methods of treating subjects having cancer utilizing the pharmaceutical compositions and dosage forms provided herein.

Pharmaceutical compositions and dosage forms

The present disclosure relates to pharmaceutical compositions comprising N-[5-(3,5-difluorobenzyl)-1H-indazol-3-yl]-4-(4-methyl-piperazin-1-yl)-2-(tetrahydro-2H-pyran-4-ylamino)-benzamide and a polymer, dosage forms comprising the formulation and methods of treating subjects having cancer.

Contemplated pharmaceutical compositions comprise a benzodiazepine anti-convulsive compound for intranasal administration in a single phase, multi-component solvent system with water content of at least 15 vol % and a benzodiazepine concentration of at least 30 mg/ml. It is noted that contrary to common expectation, the concentration of the anti-convulsive compound was increased by increasing water content in the multi-component solvent. Moreover, the anti-convulsive compound remained chemically stable and substantially completely dissolved over large periods of time. Thus, stable and highly concentrated spray formulations for intranasal administration could be prepared having sufficiently high concentrations of the anti-convulsive compounds that now allow single spray administration to achieve clinically relevant dosages.

A hydroxyalkylated heterocyclic base is prepared by reacting a heterocyclic base with an alkylene carbonate in dimethylacetamide as a solvent, wherein the hydroxyalkylated heterocyclic base is isolated from the solvent using isopropanol or tert-butylmethylether.

Infant medical hood

A scavenging medical hood includes a hood for fitting over the head of a patient. A gas inlet port leads to the interior of the hood for supplying the interior of the hood with an appropriate respiratory gas and an aerosolized medicinal agent contained within that respiratory gas. A gas outlet connects between the interior of the hood and the ambient air for discharge of gas from the interior of the hood including any residual aerosolized medicinal agent remaining in that discharged gas. A vacuum port is located adjacent to the gas outlet. A vacuum is applied to the vacuum port via a suitable motorized vacuum source. Vacuum at the vacuum port aspirates gas from the area of the gas outlet to aspirate respiratory gas, residual aerosolized medicinal agent still suspended in the respiratory gas and ambient air from the vicinity of the gas outlet into the vacuum port. A filter located in the vacuum line serves to filter out the residual aerosolized medicinal agent from the aspirate in the gas line.
