A method of making a bone matrix includes exposing a bone tissue to a solution including a surfactant and a protease; treating the bone tissue with an acid solution following exposing the bone tissue; and electrophoretically treating the acid treated bone tissue. A bone matrix has a DNA content of not greater than 0.1 micrograms per milligram sample and a modulus in a range of 180 kPa to 250 kPa.

A method of decellularizing a tissue includes disposing the tissue within a device, the device. The device includes a container, first and second electrodes disposed within the container and defining a space between the first and second electrodes to receive the tissue, a perfusion pump, and a conduit connected to the perfusion pump to transport a decellularization composition from the pump into the tissue. The method further includes disposing the decellularization composition within the container to surround the tissue and contact the first and second electrodes, applying an electric potential across the first and second electrodes, and perfusing the decullarization composition into the tissue.

The present invention additionally comprises a method and apparatus for generating and mixing signals for frequency-domain lifetime and spectral fluorometry. The present invention comprises a plurality of signal generators that generate a plurality of signals where the signal generators modulate the amplitude and/or the frequency of the signals. The present invention uses one of these signals to drive an excitation signal that the present invention then directs and transmits at a target mixture, which absorbs the energy from the excitation signal. The property of fluorescence causes the target mixture to emit an emitted signal that the present invention detects with a signal detector. The present invention uses a plurality of mixers to produce a processor reference signal and a data signal. The present invention then uses a processor to compare the processor reference signal with the data signal by analyzing the differences in the phase and the differences in the amplitude between the two signals. The processor then extracts the fluorescence lifetime and fluorescence spectrum of the emitted signal from the phase and amplitude information using a chemometric analysis.

Signal generation and mixing electronics for frequency-domain lifetime and spectral fluorometry

A method and system for measuring optical scattering characteristics includes coupling a continuous wave laser excitation signal to an optical fiber. Radiation backscattered by the optical fiber in response to the coupled excitation signal is detected to produce a backscattered radiation signal. The backscattered radiation signal is mixed with the excitation signal to produce a mixed signal. The mixed signal is filtered to reduce the magnitude of frequencies other than conjugate mixing frequencies relative to the conjugate mixing frequencies. The filtered signal is digitized and the magnitude of backscattered radiation from a specific portion of the fiber is calculated based on the digitized signal. The temperature of a specific portion of the fiber can be determined from the magnitude of the backscattered radiation.

Method and system for measuring optical scattering characteristics

There is a tremendous need for high throughput gene expression technology which can efficiently and cheaply identify and accurately isolate different genes expressed between diseased and normal tissues for use in discovering new drugs. The present invention utilizes a combination of biomolecular chemistry methods to eliminate/degrade redundant sequences and fluorescence dye assay to identify these unique sequences from two cell or tissue populations. cDNA from normal or diseased cells or tissues are hybridized with the RNA of the complement normal or diseased cells or tissues. The hybridized cDNA/RNA is incubated with exonucleases, resulting in degradation of all but the single stranded RNA and DNA. RNA are then eliminated using RNase and the remaining DNA which are unique to the sample are amplified. This technique may be used to isolate differentially expressed genes or gene fragments and will provide a means to isolate and identify medium to low gene expressions that may otherwise be buried under gene 'Noise'.

Signal generation and mixing electronics for frequency-domain lifetime

The present invention is a system for chemometric analysis for the extraction of the individual component fluorescence spectra and fluorescence lifetimes from a target mixture. The present invention combines a processor with an apparatus for generating an excitation signal to transmit at a target mixture and an apparatus for detecting the emitted signal from the target mixture. The present invention extracts the individual fluorescence spectrum and fluorescence lifetime measurements from the frequency and wavelength data acquired from the emitted signal. The present invention uses an iterative solution that first requires the initialization of several decision variables and the initial approximation determinations of intermediate matrices. The iterative solution compares the decision variables for convergence to see if further approximation determinations are necessary. If the solution converges, the present invention then determines the reduced best fit error for the analysis of the individual fluorescence lifetime and the fluorescence spectrum before extracting the individual fluorescence lifetime and fluorescence spectrum from the emitted signal of the target mixture.

Chemometric analysis for extraction of individual fluorescence spectrum

Apparatus and method for measuring the relative distance between different areas on a remote surface or displacements of a selected area of a remote surface with respect to a measuring apparatus are provided. A laser beam is modulated at an RF frequency and focused on a selected area. The selected area is scanned by movement of a mirror and at each selected area the phase difference is measured. Digital data processing of signals derived from the source and return beams is used to provide high precision. For detecting or measuring vibration or displacement, an analog signal is obtained from a low-pass filter.
