A barbecue cooking apparatus is an apparatus used to cook food items by utilizing different barbecue smoking methods. The apparatus includes a barrel, a lid, a frame, a shelved cooking system, a venting system, a winch mechanism, a lip, and a firebox gasket. The barrel provides a thermal enclosure for cooking the food items. The lid serves to close the barrel and to support the shelved cooking system. The lip and the firebox gasket seal the lid on the barrel when closed. The shelved cooking system holds both the food items within the barrel during the cooking process and the fuel source utilized to generate the heat energy to cook the food items. The frame holds the winch mechanism offset from the barrel. The winch mechanism facilitates the retrieval of the shelved cooking system from the barrel as well as the reintroduction of the shelved cooking system into the barrel.

The barrel provides a thermal enclosure for cooking the food items. The lid serves to seal the barrel during the cooking process and to support the shelved cooking system. The shelved cooking system holds the food items within the barrel during the cooking process. The shelved cooking system further holds the fuel source utilized to generate energy to cook the food items. The frame serves to hold the winch mechanism offset from the rim. The winch mechanism facilitates the retrieval of the shelved cooking system from the barrel as well as the reintroduction of the same into the barrel.

A vehicle positioning garage mat is formed as a unitary structure, including a substantially rectangular mat, with an upper surface having forward and rearward transverse bumps formed integrally thereon. The mat is to be placed on a support surface in a selected position in the path of vehicle tire, to indicate a desired parking position for the vehicle. The mat includes a rearward bump, positioned along the length of the mat forward of a trailing edge, and a forward bump spaced forwardly of the rearward bump. A tab section is provided between the rearward bump and a trailing edge of the mat for initial engagement with a vehicle tire. The tab first receives a forwardly moving vehicle tire and thus secures the mat before the tire engages the rearward bump. The forward and rearward bumps are reinforced from below by spaced longitudinal rib sections that provide resilient resistance to depression, while minimizing the amount of material required for the mat. The density of material within the mat is concentrated at the bumps such that the durometer value of the material at the bumps is greater than the overall durometer of the mat between the bumps.

Methods for the isolation and purification of the phytotoxin cercosporin are disclosed as well as methods for identifying microorganisms capable of degrading cercosporin. Cercosporin can be purified from members of the fungal genus Cercospora and incorporated into culture medium for selection of those organisms resistant to cercosporin. Once identified, these organisms can be used to isolate the protein and the gene responsible for conferring cercosporin-resistance. The gene can be cloned and inserted into a suitable expression vector so that the protein can be further characterized. Additionally, the DNA encoding for cercosporin-resistance can be inserted into a vector suitable for transforming an Agrobacterium and the Agrobacterium in turn used to transform plant cells normally susceptible to Cercospora infection. Plants can be regenerated from the transformed plant cells. In this way, a transgenic plant can be produced with the capability of degrading cercosporin.

Methods for identifying cercosporin-degrading microorganisms

The invention discloses an in vivo method for transforming and regenerating intact plants. According to the method, a plant (P) is infected with an infectious microbial agency comprised of: (1) virulence functions, (2) oncogenic factors capable of inducing a shoot-bearing shooty tumor on plant (P), and (3) a carrier vector containing engineered heterologous transfer DNA capable of being integrated into the nuclear DNA of plant (P) cells. Infected plant (P) is maintained until a shoot-bearing shooty tumor develops at or near the infection site. Those shoots, or progeny thereof, that contain transformed cells having heterologous transfer DNA integrated into their genomes are the selected and utilized to produce whole plants that contain cells having heterologous transfer DNA integrated into their genomes.
