A nanopore based sequencing system includes a plurality of nanopore sensors. Each nanopore sensor has a portion for receiving a fluid. The nanopore based sequencing system includes a fluid chamber configured to guide the fluid over the plurality of nanopore sensors and an inlet configured to deliver the fluid into the fluid chamber. At least a portion of the fluid chamber is made of a material that has been molded around at least a portion of an electrode.

Molded flow channel

A delivery system for a sensor chip includes a plurality of selectable ports and a two-way pump port selectively connectable to each of the selectable ports. The two-way pump port is configured to allow material to be drawn or delivered from or to the two-way pump port. The delivery system also includes a chamber and a bypass waste channel that is selectively connectable to the two-way pump port. The plurality of selectable ports includes a selectable chamber port connected to the chamber and the chamber has a chamber waste exit. Material may selectively flow through the chamber to a waste collection via the chamber waste exit or flow to the waste collection via the bypass waste channel that bypasses the chamber waste exit.

A nanopore cell includes a titanium nitride (TiN) counter electrode configured to be at a first electric potential. The nanopore cell also include a working electrode configured to be at a second electric potential and an insulating wall. The insulating wall and the working electrode form at least a portion of a well configured to contain an electrolyte at a voltage that is at least a portion of an electric potential difference between the first electric potential of the titanium nitride counter electrode and the second electric potential of the working electrode.

Use of titanium nitride as a counter electrode

A biochip for molecular detection and sensing is disclosed. The biochip includes a substrate. The biochip includes a plurality of discrete sites formed on the substrate having a density of greater than five hundred wells per square millimeter. Each discrete site includes sidewalls disposed on the substrate to form a well. Each discrete site includes an electrode disposed at the bottom of the well. In some embodiments, the wells are formed such that cross-talk between the wells is reduced. In some embodiments, the electrodes disposed at the bottom of the wells are organized into groups of electrodes, wherein each group of electrodes shares a common counter electrode. In some embodiments, the electrode disposed at the bottom of the well has a dedicated counter electrode. In some embodiments, surfaces of the sidewalls are silanized such that the surfaces facilitate the forming of a membrane in or adjacent to the well.

Process for biosensor well formation

A nanopore based sequencing system is disclosed. The system includes a plurality of nanopore sensors, each nanopore sensor having a top portion for receiving a fluid. The system further includes an inlet delivering the fluid into the nanopore based sequencing system and an outlet delivering the fluid out of the nanopore based sequencing system. The system includes a fluid chamber that comprises one or more fluid flow channels above top portions of the nanopore sensors; wherein the fluid chamber includes at least one divider that limits the width of the one or more fluid flow channels. In some embodiments, the at least one divider limits the width of the one or more fluid flow channel based on whether the surface tension and adhesive forces between the fluid and the fluid flow channel surfaces are sufficient to prevent the fluid from collapsing within the fluid flow channel.

Serpentine flow channels for flowing fluids over chip sensors

A delivery system for a sensor chip includes a plurality of selectable ports arranged on a first assembly. Each of the selectable ports is in communication with a separate channel. The delivery system includes a second assembly movable in relation to the first assembly. The second assembly has a channel that is mechanically connectable to different ones of the plurality of selectable ports on the first assembly by motion of the second assembly relative to the first assembly. The delivery system includes a mechanical interface configured to engage a separate actuator so that relative motion of the first assembly and the second assembly is affected by the actuator.

A nanopore based sequencing chip package is disclosed. The nanopore based sequencing chip package includes a reservoir defined by a plurality of surfaces. The chip package includes a nanopore cell array comprising a plurality of nanopore sensor cells enclosed by the reservoir. Each nanopore sensor cell has a working electrode. At least one surface of the reservoir is configured to be in contact with a conducting fluid when the conducting fluid is flowing through the reservoir. The chip package further includes a counter electrode disposed on the at least one surface of the reservoir.
