The present invention relates to the generation and use of a cellular array or a cellular array in combination with other genome-registered arrays (an array of arrays) for the determination of gene function and/or perturbation mode of action. Each cellular array consists of a number of microbial strains. Each strain comprises one reporter gene fusion made up of a gene or gene fragment operably linked to a reporter gene. Each gene or gene fragment has been &ldquo;registered&rdquo; or mapped to a specific location in the genome of the organism. The genome-registered collection of the invention may be used to determine alterations in gene expression under a variety of conditions. Such collections are amenable to rapid assay and may be used to confirm, correct or augment data generated from DNA micro array technology.

Cellular arrays for the identification of altered gene expression

The global effect on genes under different environmental conditions can be determined by a comprehensive gene expression profile. The present invention provides a method to monitor the changes in comprehensive cellular gene expression levels at single length resolution by using a high-density microarray prepared with a comprehensive collection of ORFs of a genome. Under different environmental conditions, directly and indirectly affected genes can be detected as the gene expression levels are induced or repressed in comparison to the control.

Method for high-density microarray medicated gene expression profiling

A new method for the identification of useful promoters is disclosed. The method is capable of identifying bacterial promoters sensitive to a particular cellular insult and may be modified to identify promoters sensitive to herbicides and crop protection chemicals. Constructs comprising promoters upstream of a luminescent reporter genes are placed in transformed hosts. Transformants grown in liquid media to a predetermined growth stage and contacted with a cellular insult are assessed for regulatory region activity by measurement of the resulting change in bioluminescence. The method is able to identify promoters undetectable by standard methods.
