Graphitic nanotubes, which include tubular fullerenes (commonly called 'buckytubes') and fibrils, which are functionalized by chemical substitution, are used as solid supports in electrogenerated chemiluminescence assays. The graphitic nanotubes are chemically modified with functional group biomolecules prior to use in an assay. Association of electrochemiluminescent ruthenium complexes with the functional group biomolecule-modified nanotubes permits detection of molecules including nucleic acids, antigens, enzymes, and enzyme substrates by multiple formats.

Graphitic nanotubes in luminescence assays

An apparatus for performing a binding assay for an analyte of interest present in a sample based upon measurement of electrochemiluminescence at an electrode surface comprising a cell defining a sample containing volume intersecting with inlet and outlet means, an electrode having a substantially horizontally positioned surface exposed to and positioned below a portion of the sample containing volume, means for impressing electrochemical energy upon said electrode sufficient to generate luminescence, means for magnetically collecting particles along said surface and means for measuring the luminescence generated at said electrode.

Methods and apparatus for improved luminescence assays

A method for increasing the rate of chemical reactions involving the conversion of at least one reactant to at least one product involves contacting the reactant with an appropriate monoclonal antibody under conditions permitting the formation of a complex between the antibody and the reactant. The complexed reactant is converted to the product which is then released from the complex. The monoclonal antibody may employ a cofactor and may be directed to a known substrate of an enzyme. Methods for preparing such monoclonal antibodies are also disclosed.

Catalytic antibodies

Antigens capable of eliciting antibodies which can catalyze chemical reactions, in particular, the cleavage or formation of a peptide linkage, comprising a hapten or a hapten and a suitable carrier molecule are disclosed. Haptens include, among others, silicon and boron containing compounds. Antibodies which are catalytically active for chemical reactions, in particular, the cleavage or formation of a selected peptide linkage or an ester bond, and which are elicited by such antigens are disclosed as well as methods for producing the antibodies and methods for catalyzing the cleavage or formation of a peptide linkage or in ester bond in a molecule.

Transition state analogs

Qualitative and quantitative electrochemiluminescent assays for analytes of interest present in multicomponent liquids are provided. These methods comprise contacting a sample with a reagent labeled with an electrochemiluminescent chemical moiety and capable of combining with the analyte of interest, exposing the resulting sample to electrochemical energy and detecting electromagnetic radiation emitted by the electrochemiluminescent chemical moiety. Further provided are methods for detecting and identifying the presence of a multiplicity of analytes in a liquid food or food homogenate. These methods comprise immersing a diagnostic reagent holder, provided with a multiplicity of reagents, into the food or food homogenate, removing the diagnostic reagent holder from the liquid food or food homogenate, and detecting and identifying the presence of a multiplicity of analytes of interest bound to the diagnostic reagent holder, thereby detecting and identifying the presence of a multiplicity of analytes of interest in the food or food homogenate. The invention further provides an enzyme immunoassay for coliform bacteria. This assay comprises inoculating a sample into a suitable medium for coliform reproduction, immobilizing coliforms present in the medium to a suitable surface, treating the surface with an antibody directed to the immobilized coliforms and detecting the presence of the immobilized coliforms immobilized to a suitable surface.

Electrochemiluminescent assays

The invention provides compositions and kits for performing a binding assay for an analyte of interest present in a sample based upon electrochemiluminescence. The compositions and kits comprise an electrochemiluminescent label, collectable particles, binding reagents, and an electrolyte.

A system for detecting physical or chemical changes in an environment is disclosed. The system comprises means for conducting a chemical reaction; an active biological molecule whose physiological function is noncatalytic which is capable of catalyzing a chemical reaction wherein at least one reactant is converted to one product; and means for sensing and processing information relating to changes in said environment. A method for detecting physical or chemical changes in an environment is also disclosed.

Catalytic antibodies as chemical sensors

Catalytic antibodies are disclosed. The catalytic antibodies are specific for prodrug compounds. The catalytic antibodies enhance cleavage of an active drug moiety from a prodrug residue, thereby activating the drug.

Prodrugs activated by targeted catalytic proteins

Antigens capable of eliciting antibodies which can enhance the rate of chemical reactions at peptide bonds are disclosed. In particular, the rate of cleavage or formation of metastable peptide bonds, such as ASN-X, ASP-X, GLN-X, GLU-X, LYS-X, and HIS-Y-X, where X and Y are any amino acid, is enhanced by antibodies elicited by said antigen.
