The present invention relates to excision of explant material comprising meristematic tissue from cotton seeds. Methods for tissue preparation, storage, transformation, and selection or identification of transformed plants are disclosed, as are transformable meristem tissues and plants produced by such methods, and apparati for tissue preparation.

A method for depositing particles coated with biological substances onto the concave inner surface of a length of tubing includes the steps of preparing a uniform suspension of coated particles, introducing the particles into the tubing, placing the tubing into a generally horizontal position, removing the evaporable liquid after the particles have settled, and drying the particles. When dry, the tubing is cut into sample cartridges of appropriate length for use in a particle acceleration instrument. An apparatus for performing the method is also disclosed.

An apparatus for acceleration of carrier particles into a target organism which may be used for rapid, sequential particle acceleration procedures comprises a holder for removable mounting cartridges. A first removable mounting cartridge detachably holds a carrier sheet coated with carrier particles which have themselves been coated with biological material. A second removable mounting cartridge comprises a retaining screen. The mounting cartridges may be inserted into the holder before particle acceleration and replaced quickly afterwards, thereby readying the apparatus for another particle acceleration. Convenient replacement of these consumable items eliminates the need to disassemble the apparatus between each particle acceleration procedure.
