A microscope includes a holder for holding a sample, an objective for imaging at least apart of a sample held by the holder, a detection module, a control unit for setting the focus position of the objective in a first direction for the recording by means of the detection module, and a focusing module for maintaining a set focus position of the objective. The focusing module includes the control unit, a second detector and first focusing optics with adjustable focal length. The focusing module is switchable into a focus-hold mode, wherein an intensity-modulated object is imaged into the sample via the first focusing optics and the objective, and an image of the imaged object is recorded by means of the second detector. The control unit holds the focus position of the objective on the set focus position, based upon the recording of the second detector.

A microscope including an objective having a focal plane in a sample space, and an autofocus device comprising a light modulator for generating a luminous modulation object that is intensity-modulated periodically along one direction, an autofocus illumination optical unit that images the modulation object such that its image arises in the sample space, an autofocus camera, an autofocus imaging optical unit that images the image of the modulation object in the sample space onto the autofocus camera, a control device, which receives signals of the autofocus camera and determines an intensity distribution of the image of the modulation object and generates a focus control signal therefrom. The control device determines an intensity distribution of the image of a luminous comparison object imaged by the optical unit to correct the intensity distribution of the image of the modulation object with regard to reflectivity variations in the sample space.

Autofocus method for microscope and microscope comprising autofocus device

A method for detecting and controlling supply of an immersion medium into an immersion film region between the front lens of a microscope objective and a preparation, particularly in automated microscopes, using a light source which is reflected into the microscope beam path by means of a beam splitter. The state of the immersion film region is recorded and evaluated in a detection unit of the microscope or a detection unit of the autofocus device through an autofocus signal of the light source and/or a reflection of a modulation object arranged in the intermediate image plane of an incident light illumination, and is transmitted to a control unit for the purpose of a change or an error treatment of the immersion process.

Method for detecting and controlling supply of an immersion medium

A method is provided for determining the distance between two optical boundary surfaces spaced apart from each other in a first direction. A first image is ascertained wherein the plane into which the pattern acquired coincides with a first of two optical boundary surfaces or has the smallest distance to the first optical boundary surface in a first direction. A position of the first image in the first direction is determined. A second image is ascertained wherein the plane into which the pattern acquired coincides with a second of two optical boundary surfaces or has the smallest distance to the second optical boundary surface in the first direction. The position of the second image in the first direction is determined. The distance is calculated by means of determined positions of the first and second image.

A procedure and a device for terminating the immersion at a microscope having one or more immersion lenses, or both immersion lenses and dry lenses, comprising a protective sheath around the immersion film area for preventing the escape of immersion media and an immersion media supply unit and an immersion media discharge unit connected with the protective sheath. The immersion media is removed in a controlled manner in particular in an automated microscope. Subsequently, the microscopic testing can be continued without the user having to intervene in the device or the process.

Procedure and device for terminating the immersion at a microscope

An autofocus method for a microscope with an objective which images a sample lying in an object plane, including the steps: projecting a longitudinally extended grating slit which lies in a grating slit plane onto the sample, and imaging the projection of the grating slit onto an autofocus camera; determining an intensity distribution of the grating slit image and from this, deducing a preset for a relative adjustment of sample and object plane; projecting a likewise longitudinally extended comparison slit onto the sample, and imaging the projection of the comparison slit onto the autofocus camera; evaluating the width of the comparison slit image at right angles to the longitudinal extension at at least two sites which are spaced apart along the longitudinal extension, and determining a width variation of the comparison slit image, a gradient of the width variation and a direction of the relative adjustment.

A microscope including an imaging optical unit, a sample stage for supporting a sample to be examined, a movement unit, by which the distance between sample stage and imaging optical unit can be altered, a focus measuring unit, which measures the present focus position and outputs a focus measurement signal, a control unit for maintaining a predetermined focus position for examinations of the sample that are separated from one another in time. The control unit receives the focus measurement signal and derives a deviation of the present focus position from the predetermined focus position. Dependent on the deviation derived the movement unit, changes the distance between sample stage and imaging optical unit so that the predetermined focus position is maintained. The control unit drives the movement unit () for maintaining the predetermined focus position only before and/or after at least one of the examinations, but never during the examinations.

A microscope including an objective, which images a sample along a microscope beam path, and an autofocus device, which is coupled into the microscope beam path via a beam splitter at a location behind the objective. A light modulator for generating a two-dimensional, intensity-modulated modulation object, is located in the autofocus beam path in a plane conjugated to the focal plane of the objective or intersects the latter and is imaged into the focal plane of the objective. A camera records a two-dimensional image onto which the modulation object's image is imaged. The image plane of the camera intersects a plane that is conjugated to the modulation object or is located in the plane and the camera detecting the contrast of the modulation object's image located in the sample.

The invention relates to a microscope having a stage for supporting a sample to be examined, a recording sensor, an imaging optic for imaging the sample onto the recording sensor, a moving unit by means of which the distance between the stage and the imaging optic can be changed, a control unit for controlling an image recording of the sample and a focus-holding unit for maintaining a prescribed focal position for image recording of the sample at temporal intervals, wherein the focus-holding device comprises at least one hardware element and one software module, wherein the focus-holding unit is fully integrated in the control unit, on both the hardware and software sides.

The invention relates to an optics changer for arranging an optical element in a target position in a changer chamber of an optical device accessible from the outside via a insertion channel, including a base frame and the optical element that is pivotally fastened to the base frame via a swivel mechanism, wherein the swivel mechanism effects a pivoting movement of the optical element when the optics changer is inserted through the insertion channel in the changer chamber starting at a predetermined insertion depth such that after inserting, the optical element is placed and pivoted in the target position.

Fluorescence microscope with blocking filters for a portion of the light emitted by a specimen have markings referring to the orientation of their wedge angle. A marking to which the marking on the filter can be oriented in a defined manner is preferably provided on the filter holder of the microscope, the filters being marked with respect to their wedge angle.
