Chromatographic systems for size exclusion chromatography (SEC) are provided that comprise an inlet, an outlet, an analytic column having a first interior volume that has a first length and a first cross-sectional area normal to the first length, the first interior volume containing a first stationary phase, and a guard column having a second interior volume that has a second length and a second cross-sectional area normal to the second length, the second interior volume containing a second stationary phase. The inlet is in fluid communication with the guard column, the guard column is in fluid communication with the analytic column, and the analytic column is in fluid communication with the outlet. Moreover, the second length is smaller than the first length, and the second cross-sectional area is smaller than the first cross-sectional area.

Guard column configurations for size exclusion chromatography separations

The present disclosure pertains to sample preparation devices useful for affinity capture and purification that include one or more internal structures that comprise a reservoir, a well, a fluid passageway, sorbent particles, and a filter element that blocks passage of the affinity sorbent particles, which sample preparation devices combine the attributes of both dispersive and flow through designs into a single sample preparation device. The present disclosure also pertains to kits that contain and methods that use such sample preparation devices.

This disclosure pertains to performance enhancing conditioning and storage solvents containing low levels of buffer and salt for reproducible and improved size exclusion chromatography (SEC). In some embodiments, the present disclosure pertains to storage solvents for protein-based size exclusion chromatography that include water, a water-miscible organic solvent, a phosphate salt, and a neutral salt. In some embodiments, the present disclosure pertains to columns for protein-based size exclusion chromatography that include porous particles and such a storage solvent.

Exemplary embodiments are directed to devices for separating a sample by chromatography, components of the devices, and methods for using the devices, and directed to devices and components for use with immobilized enzymatic reactors. A device includes a wall having a wetted surface exposed to a mobile phase including the sample during chromatographic separation. The wetted surface of the wall includes an alloy material including the following constituents: nickel, and cobalt and/or chromium where the alloy is limited in an amount of titanium to 1 wt %. A component includes a body having a wetted surface exposed to a mobile phase including the sample during chromatographic separation. The wetted surface of the body includes an alloy material including the following constituents: nickel, and cobalt and/or chromium where the alloy is limited in an amount of titanium to 1 wt %.

A clamp assembly includes a rail configured to receive a first fluidic assembly, and a carriage slidably mounted to the rail and configured to receive a second fluidic assembly. The carriage is operable to establish a first fluid tight seal between the first fluidic assembly and a chromatography column received within the claim assembly, and to establish a second fluid tight seal between the second fluidic assembly and the chromatography column.

The invention provides filters and methods of assembling filters. In an exemplary embodiment, the filter includes a porous element, a compression element, and a housing. The compression element can be configured to receive the porous element, thereby forming an assembly. For example, the compression element can receive the porous element in a slip-fit relationship. The housing can have an opening formed therein that is configured to receive the assembly. In some embodiments, the assembly can be retained within the opening when the assembly is received therein. For example, the opening can receive the assembly in a press-fit relationship.

Described is a chromatographic column assembly that includes an outer tube comprising a metal, a first conduit disposed within the outer tube, a second conduit disposed within the outer tube, and a first joint located between the first conduit and the second conduit. The outer tube is deformed by a first uniform radial crimp at a longitudinal location along the outer tube that surrounds the first conduit on a first side of the first joint, and a second uniform radial cramp at a longitudinal location along the outer tube that surrounds the second conduit on a second side of the first joint. The first and second uniform radial cramps form a fluid-tight seal between the first conduit and the second conduit and each have a substantially flat base region over which a diameter of the outer tube is reduced for a non-zero longitudinal length.

Methods, compositions, devices, and kits, for performing phospholipid separation

Described is a chromatography column assembly that includes a permanently deformable outer tube, an intermediate tube, an inner tube and a sorbent bed disposed within the inner tube. The sorbent bed may be in the form of packed chromatographic particles or a porous monolithic structure. A radial seal is provided by one or more uniform radial crimps at longitudinal locations on the assembly. The uniform radial crimp compresses the outer tube and underlying intermediate tube onto the inner tube to achieve a high pressure liquid tight seal between the three tubes. The length and depth of each crimp is accurately formed to accommodate the requirements of the particular application. Leakage along the tubing assembly is prevented and void volume is reduced or eliminated. No external ferrule or ferrule swaging mechanism is needed; therefore the chromatography column assembly is easily adapted for use in various chromatographic column configurations.

Methods, compositions, devices, and kits for performing phospholipid separation

A frit () includes a porous support structure having a plurality of void spaces, where a plurality of secondary particles () are filled in the void spaces, the secondary particles () being dimensioned with respect to the void spaces such that the frit () retains packing materials with diameters of less than about 2.5 microns. Preferably one or more frits () are received in a high pressure liquid chromatography (HPLC) chromatographic column, where the column includes fittings and filter assemblies for receiving frits () at its inlet and outlet. The frit () can be constructed from a porous support structure with void spaces or pores that are filled with secondary particles () smaller those used to manufacture the support. The particles () contained in the void spaces are large enough to be retained by the support, but small enough to create a finer network of interconnected channels within the support's void spaces that are capable of retaining sub-2.5 micrometer particles.
